Reference map of the low molecular mass proteins of Haemophilus influenzae

Abstract: Analysis of the proteome of Haemophilus influenzae by two-dimensional polyacrylamide gel electrophoresis on conventional Tris-glycine gels does not usually result in efficient separation of the proteins in the 5-20 kDa range, which are mainly accumulated in the lower acidic and basic regions. In order to improve the separation of the low molecular mass proteins, we used homogeneous Tricine gels of two urea concentrations in the second-dimensional separation. The Tricine gel systems allowed the efficient and re… Show more

“…The 2D-electrophoresis was adapted to the separation of proteins with low molecular mass (Fountoulakis et al, 1998). Identification of proteins from excised spots was carried out by analysis of the peptide mixtures from tryptic digests (Perspective Biosystems, Framingham, MA, USA), using the Voyager-DE STR device and Profound software (prowl.rockfeller.edu/cgi-bin/Profound).…”

“…The vapBC alleles in NTHi have been shown to be expressed by proteomic studies (12,17,19). As NTHi carries two chromosomal vapBC operons (HI0321-HI0322 and HI0948-HI0947), for clarity we shall refer to these as vapBC-1 and vapBC-2.…”

“…In order to analyze comprehensively all "natural" peptides and small proteins of a whole organism (peptidome), an approach described as peptidomics is used (28). Neither the 2D gel approach (which is limited to proteins Ͼ10 kDa (29)) nor the shotgun approach can therefore be used to analyze the naturally occurring peptides. The workflow of peptidomics usually starts with the removal of the proteins over 20 kDa by size-exclusion filtration or size fractionation with membranes of suitable pore size.…”
