Reference gene selection for quantitative PCR in liver, skeletal muscle, and jejunum of Bos indicus cattle

Abstract: The objective of the present study was to evaluate the stability of candidate reference genes and select the genes that can be used for normalizing realtime polymerase chain reaction (PCR) in the liver, skeletal muscle, and jejunum tissues of Nellore or Nellore × Angus steers fed different diets. Fourteen purebred and 14 crossbred steers were used, in which half of the animals of each genetic group received a diet containing whole shelled corn (WSC) and the other half whole shelled corn and sugarcane bagasse (… Show more

“…RT-qPCR reactions was carried out using PowerUp SYBR Green Master Mix (Applied Biosystems), with 40 amplification cycles at 60˚C. The standard curve method was applied to calculate relative gene expression using GAPDH, RPL0, and ACTB as internal control [33,[35][36][37]. The mean of these genes for each sample was taken to provide an optimum basis for a normalization of the target genes [38,39].…”

“…The relative mRNA expression was determined using the threshold cycle (CT) method, using the primers of each gene. The β-actin and GAPDH ( D-glyceraldehyde-3-phosphate dehydrogenase ) were used as reference genes for an expression analysis for liver samples, while β-actin and CASC3 ( Cancer susceptibility candidate 3 ) were used as reference genes for skeletal muscle samples [ 6 , 31 ] samples.…”

“…The algorithms used in this study, geNorm 59 , NormFinder 62 , BestKeeper 63 , Delta-Ct 64 , and RefFinder 65,66 have been commonly used to evaluate the stability of reference genes in plants 19,25,54 , animals [67][68][69] and microorganisms [70][71][72] . They are used to calculate the stability of gene expression using different mathematical models.…”
