Reductive Dehalogenation of Chlorobenzene Congeners in Cell Extracts of Dehalococcoides sp. Strain CBDB1

Abstract: Enzymatic reductive dehalogenation of tri-, tetra-, penta-, and hexachlorobenzenes was demonstrated in cell extracts with low protein concentration (0.5 to 1 g of protein/ml) derived from the chlorobenzene-respiring anaerobe Dehalococcoides sp. strain CBDB1. 1,2,3-trichlorobenzene dehalogenase activity was associated with the membrane fraction. Light-reversible inhibition by alkyl iodides indicated the presence of a corrinoid cofactor.

“…Reductive dechlorination of chlorinated benzenes and PCDDs which had previously been shown for strain CBDB1 (2,9,18) was also recently demonstrated for strain 195 (11). However, different patterns of chlorobenzene dechlorination (3) and differences in the utilization of chlorobenzene and PCDD congeners have been observed for strain CBDB1 (2,9,17,18) and strain 195 (11). For example, hexachlorobenzene (HCB) dechlorination by strain 195 mainly follows the pattern HCB3pentachlorobenzene (PeCB)31,2,3,5-tetrachlorobenzene (TeCB)31,3,5-trichlorobenzene (TCB) (11) that was also described for the Dehalococcoides-like bacterium DF-1 identified in a mixed culture (50).…”

In contrast, HCB dechlorination in strain CBDB1 proceeds via the two isomers 1,2,4,5-TeCB and 1,2,3,5-TeCB and leads to the formation of dichlorobenzenes and 1,3,5-TCB (17,18). In contrast to strain 195 and bacterium DF-1, strain CBDB1 is able to grow with 1,2,3-TCB and 1,2,4-TCB as the sole electron acceptors (2).…”

“…Most RDases contain a corrinoid cofactor and 2 ironsulfur clusters. In fact, the RDase activity of most dehalorespiring species is light-reversibly inhibited by alkyl iodides, indicating the involvement of corrinoids as a cofactor 69,86) . The rdh genes are mostly adjacent to the genes for transcriptional regulators such as two-component regulatory systems or transcriptional regulators of the MarR-type, indicating that the expression of the rdhAB genes is tightly regulated.…”

“…The N termini of RDases contain a twin-arginine signal sequence with the consensus motif RRXFXX, which is involved in transporting cofactor-containing proteins across the cytoplasmic membrane. Actually, TCE-RDase of "Dehalococcoides ethenogenes" 85) , the chlorobenzeneRDase of "Dehalococcoides ethenogenes" strain CBDB1 6,69) , and PceA-RDase of Desulfitobacterium hafniense strain Y51 121) are located on the exterior of the cytoplasmic membrane. Most RDases contain a corrinoid cofactor and 2 ironsulfur clusters.…”

“…obtained in pure culture, strain CBDB1, (98.3% identity over the 16S rRNA gene sequence of strain 195 and CBDB1), was the first reported isolate able to obtain energy from chlorobenzene reductive dechlorination (Adrian et al, 2000;Hölscher et al, 2003;Jayachandran et al, 2003). Strain CBDB1 can also reductively dechlorinate selected chlorinated dibenzo-p-dioxins (Bunge et al, 2003) and chlorophenol congeners .…”
