Reduction of Covalent Binding of Aflatoxin B1 to Rabbit Liver DNA After Immunization Against this Carcinogen

Abstract: Abstract. The covalent binding of [ 3 H]aflatoxin B 1 (AF) to liver DNA was determined, 6 h after oral administration to male rabbits. A Covalent Binding Index, CBI (flmol AF/mol DNA-P)/(mmol AF/kg b. w.) = 8,500was found. Pretreatment of rabbits with AF coupled to bovine serum albumin in Freund's adjuvant led to the production of AF-directed antibodies. Administration of [ 3 H]AF to such immunized rabbits resulted in a CJH of only 2,500, i.e., the iiDJ{.lUnization provided a protection by a factor of more tha… Show more

“…The high antibody titer in occupationally exposed individuals may be due to concomitant exposure to endotoxins that act as an adjuvant. Antibodies aginst a carcinogen epitope have been shown to decrease the uptake of the carcinogen after oral administration (11), inhibit carcinogen-DNA binding (12), and inhibit tumorigenicity (13). Dietary exposure to aflatoxins alters the immune function (14), but the biological consequence of antibodies against AFB remains unknown.…”

“…Lower mortality and reduction of acute toxic effects in the liver were also demonstrated in immunized rabbits and rats challenged with a single dose of AFB 1 [ 120 , 121 , 122 , 123 ]. Following similar immunization experiments, a reduction of the covalent binding of AFB 1 to liver DNA was observed, indicating a possible reduced susceptibility for liver tumor formation [ 124 ]. Additional in vitro investigations proved that specific anti-AFB 1 antisera were able to decrease genotoxic and mutagenic effects of AFB 1 [ 125 , 126 ].…”
