Reducing the nicotine content of tobacco by grafting with eggplant

Abstract: Background: Nicotine is a stimulant and potent parasympathomimetic alkaloid that accounts for 96-98% of alkaloid content. A reduction in the amount of nicotine in cigarettes to achieve a non-addictive level is necessary. We investigated whether replacing tobacco root with eggplant by grafting can restrict nicotine biosynthesis and produce tobacco leaves with ultra-low nicotine content, and analyzed the gene expression differences induced by eggplant grafting. Results: The nicotine levels of grafted tobacco lea… Show more

“…The pathways related to the biosynthesis of amino acids and the metabolism of various amino acids have been indicated to be responsible for the degradation of TSNAs [4,7]. In addition, the metabolic 10/18 process of amino acids could reportedly reduce the production of TSNAs by reducing the levels of nitrite [4].…”

“…The metabolite data highlighted a negative correlation of nicotine to precursors aspartic acid, cadaverine and glutamic acid as well as amino acids glutamine, GABA, threonine, serine, alanine and glycine and polyamine spermidine. Reduction of nicotine content has been previously associated with higher content of precursors (Ren et al, 2020). This implies that more assimilated nitrogen is available for amino acids and subsequently protein biosynthesis in Nicotiana plants with reduced pyridine alkaloid biosynthesis.…”

“…Grafting is a widely used agronomic practice that can be applied to improve tobacco yield (Ren et al, 2020 ). Nicotine is synthesized in tobacco roots and transported to leaves, and it has previously been found that the leaves of tobacco grafted onto a tomato rootstock were characterized by no significant accumulation of nicotine (Dawson, 1942 ).…”

Similarly, the leaves of tobacco that had been grafted onto an eggplant rootstock were found to contain very low levels of nicotine (Ren et al, 2020 ).…”
