Reduced oxygen concentration during in vitro oocyte maturation alters global DNA methylation in the maternal pronucleus of subsequent zygotes in cattle

Abstract: Preimplantation epigenetic reprogramming is sensitive to the environment of the gametes and the embryo. In vitro maturation (IVM) of bovine oocytes is a critical step of embryo in vitro production procedures and several factors influence its efficiency, including atmospheric oxygen tension. The possibility that the IVM environment can alter this process is tested by determining whether the global DNA methylation pattern (measured via immunofluorescent labeling of 5-methylcytosine [5meC]) in the parental pronuc… Show more

“…These results indicated that 5% CO 2 + 5% O 2 support the development of in vitro produced buffalo embryos to the blastocyst stage better than 20% O 2 conditions. These results are concomitant with those previously reported in cattle [9,23] and pigs [24]. Embryos cultured under OS showed higher ROS accumulation and higher abundance of transcripts related to OS response, which, in turn, decreases cell proliferation and number.…”

“…In addition, oxygen concentrations might have an effect on the epigenetic reprogramming of oocytes and preimplantation embryos. For example, a recent study reported a significant difference of 5-methylcytosine between maternal pronuclei of bovine oocytes matured at 5% and 20% oxygen (Bennemann et al, 2018). However, literature on the relationship between oxygen tension and epigenetic status are scarce and their long-term sequences still needs to be further investigated.…”

“…However, literature on the relationship between oxygen tension and epigenetic status are scarce and their long-term sequences still needs to be further investigated. What is more, it must be mentioned that the actual dissolved oxygen concentration during IVM and IVC is generally lower than that in the incubator as the solubility of oxygen is poor in the culture medium (Bennemann et al, 2018;Talevi et al, 2018). The dissolved oxygen concentration is also affected by multiple factors such as the volume of the drop of culture medium, the amount of mineral oil applied, and the culture procedures (Sciorio and Smith, 2019).…”

“…In vivo maturation. COCs were washed in 3 subsequent 100 μl drops of holding medium (0.453 g TCM199 supplemented with 1.5 mg gentamicin sulphate; 0.66 mg sodium pyruvate; 0.03 g NaHCO 3 ; 0,03 g BSA fatty acid free; dissolved in 30 ml sterile water) 75 . The transfer of COCs was conducted in a standardized volume using a Stripper micropipettor (The Stripper, MXL3-135, Origio a/s, Måløv, Denmark) adjusted to 2.5 μl.…”

“…In vitro maturation. Collected immature COCs were washed individually in four consecutive 100 μl drops of maturation wash medium (0.453 g TCM199 supplemented with 1.5 mg gentamicin sulphate; 0.66 mg sodium pyruvate, 0.066 g NaHCO 3 ; 0.03 g BSA fatty acid free; dissolved in 30 ml sterile water) and transferred to 30 μl drops maturation medium (supplemented with 10 IU/ml equine chorionic gonadotrophin and 5 IU/ml human chorionic gonadotrophin) 75 . In vitro maturation was performed for 21 h in individual culture at 38.5 °C and 5% CO 2 .…”
