At high concentrations, free radicals and radical-derived, nonradical reactive species are hazardous for living organisms and damage all major cellular constituents. At moderate concentrations, however, nitric oxide (NO), superoxide anion, and related reactive oxygen species (ROS) play an important role as regulatory mediators in signaling processes. Many of the ROS-mediated responses actually protect the cells against oxidative stress and reestablish “redox homeostasis.” Higher organisms, however, have evolved the use of NO and ROS also as signaling molecules for other physiological functions. These include regulation of vascular tone, monitoring of oxygen tension in the control of ventilation and erythropoietin production, and signal transduction from membrane receptors in various physiological processes. NO and ROS are typically generated in these cases by tightly regulated enzymes such as NO synthase (NOS) and NAD(P)H oxidase isoforms, respectively. In a given signaling protein, oxidative attack induces either a loss of function, a gain of function, or a switch to a different function. Excessive amounts of ROS may arise either from excessive stimulation of NAD(P)H oxidases or from less well-regulated sources such as the mitochondrial electron-transport chain. In mitochondria, ROS are generated as undesirable side products of the oxidative energy metabolism. An excessive and/or sustained increase in ROS production has been implicated in the pathogenesis of cancer, diabetes mellitus, atherosclerosis, neurodegenerative diseases, rheumatoid arthritis, ischemia/reperfusion injury, obstructive sleep apnea, and other diseases. In addition, free radicals have been implicated in the mechanism of senescence. That the process of aging may result, at least in part, from radical-mediated oxidative damage was proposed more than 40 years ago by Harman ( J Gerontol 11: 298–300, 1956). There is growing evidence that aging involves, in addition, progressive changes in free radical-mediated regulatory processes that result in altered gene expression.

Σκοπός: Σκοπός της μελέτης ήταν η μεταβολή της οξειδοαναγωγικής κατάστασης στομυϊκό ιστό μέσω της χορήγησης NAC, μετά από ασκησιογενή μυϊκό τραυματισμό και ηεξέταση των επιπτώσεων αυτής της μεταβολής στη λειτουργική κατάσταση του μυ, στηνεκδήλωση της φλεγμονής και στην πρωτεϊνική έκφραση των βασικότερων πρωτεϊνών πουχαρακτηρίζουν τον κύκλο της ασκησιογενούς φλεγμονής του μυϊκού κυττάρου. Μέθοδος:Δέκα απροπόνητοι άντρες εκτέλεσαν δυο πειραματικές συνθήκες όπου στη μια λάμβανανNAC και στην άλλη Placebo. Η κάθε συνθήκη έγινε σε διαφορετικό πόδι καιπραγματοποιήθηκαν 300 έκκεντρες μυϊκές συστολές σε ισοκινητικό δυναμόμετρο.Δείγματα αίματος λήφθηκαν πριν και αμέσως μετά την άσκηση καθώς και στις 2h, 1d, 2d,3d, 4d, 5d, 6d, 7d και 8d μετά την άσκηση. Στα ίδια χρονικά σημεία έγινε εκτίμηση τηςαπόδοσης και του μυϊκού τραυματισμού. Μυϊκές βιοψίες λήφθηκαν πριν την άσκηση, στις2h, στις 2d και στις 8d μετά την άσκηση. Δείκτες που μετρήθηκαν: Για τηνοξειδοαναγωγική κατάσταση στον μυ η GSH, η GSSG και ο λόγος τους GSH/GSSG. Γιατο οξειδωτικό στρες στο μυ τα TBARS και τα PC. Για τη μυϊκή απόδοση στις 60ο/sec ημέγιστη ροπή, η σχετική μέγιστη ροπή, η μέση ροπή και η σχετική μέση ροπή. Για τονμυϊκό τραυματισμό τα DOMS, το KJRM και η CK. Για τη φλεγμονή τα WBC, η CRP, ηIL-1β, η IL-6, η IL-10, ο ICAM-1, ο VCAM-1, η Σελεκτίνη P, η Σελεκτίνη E και ηδιείσδυση των μακροφάγων στο μυϊκό ιστό. Για τα μεταγωγικά μονοπάτια ηφωσφορυλίωση των πρωτεϊνών p-PKB, p-mTOR, p-p70S6k, MyoD, p38, p-NF-kB, prpS6,TNF-a. Αποτελέσματα: Η NAC μετέβαλε την οξειδοαναγωγική κατάσταση στο μυκαι τον προστάτεψε από το οξειδωτικό στρες. Στη συνθήκη NAC εμφανίστηκε μικρότερηπτώση της απόδοσης αλλά και καθυστερημένη επαναφορά της. Στους δείκτες μυϊκούτραυματισμού μόνο η CK περιορίστηκε μετά τη λήψη της NAC. Οι περισσότεροι δείκτεςφλεγμονής φάνηκε να περιορίζονται με τη λήψη του αντιοξειδωτικού. Τέλος η NACφάνηκε να επιδρά αρνητικά στα μεταγωγικά μονοπάτια που σχετίζονται με την επούλωσητου μυ. Συμπέρασμα: Η NAC μπορεί να περιορίσει την εμφάνιση της φλεγμονής ωστόσο θα καθυστερήσει την επαναφορά της απόδοσης καιενδεχομένως την επούλωση του μυϊκού ιστού.

Effect of Oxidized Fibrinogen on Aggregation of Activated Platelets and Neutrophils

The effect of oxidized fibrinogen on platelet-neutrophil complex formation was evaluated by studying the platelet aggregation (changes in light transmission and turbidimetric assay). Activation of cells by thrombin (0.015 U/ml) in the presence of oxidized fibrinogen was accompanied by the formation of larger intermolecular aggregates of platelets and leukocytes as compared to those detected in experiments with non-oxidized fibrinogen. Addition of thrombin (0.2 U/ml) in the presence of oxidized fibrinogen was followed by the formation of more stable complexes of platelets and leukocytes as compared to those revealed in experiments with non-oxidized fibrinogen. An increase in the width of aggregation curves was most pronounced in the system of 10(-4) M Fe(2+) and 10(-4) M H(2)O(2) with oxidized fibrinogen. Our results indicate that oxidized fibrinogen contributes to the "floating" or suspension of platelet-leukocyte complexes.
