Redox control of protein degradation

Abstract: Intracellular proteolysis is critical to maintain timely degradation of altered proteins including oxidized proteins. This review attempts to summarize the most relevant findings about oxidant protein modification, as well as the impact of reactive oxygen species on the proteolytic systems that regulate cell response to an oxidant environment: the ubiquitin-proteasome system (UPS), autophagy and the unfolded protein response (UPR). In the presence of an oxidant environment, these systems are critical to ensure… Show more

“…. Recently, NRF2 was also found to regulate the expression of several proteasome subunits and autophagy genes, providing additional interest for its control of proteostasis (Pajares et al, 2015(Pajares et al, , 2016(Pajares et al, , 2017de la Vega et al, 2016).…”

“…Although numerous reports establish a role for ROS in stimulating autophagy, the mechanisms by which specific ROS promote autophagy remains unclear [54, 59, 60]. Nonetheless, it appears that both superoxide and H 2 O 2 can accelerate autophagy in several ways.…”

“…Bearing in mind that oxidized proteins accumulate in aging and in several age-related pathological conditions [23], when CMA activity has been shown to decline [24,25], this study provides a new strategy to activate CMA through NFE2L2 inducers to facilitate elimination of such damaged and otherwise toxic proteins.…”
