Red wine polyphenols prevent angiotensin II-induced hypertension and endothelial dysfunction in rats: Role of NADPH oxidase

Abstract: Intake of red wine polyphenols prevents Ang II-induced hypertension and endothelial dysfunction. Prevention of vascular NADPH oxidase induction and preservation of arterial nitric oxide availability during Ang II administration likely contribute to this effect.

“…The NO-mediated component of relaxation was unaltered in mesenteric arteries from angiotensin II-treated rats. In contrast, in large elastic arteries such as aorta in which NO is the main endothelium-derived relaxing factor, chronic administration of angiotensin II induced a significant inhibition of NO-mediated relaxation (Sarr et al, 2006). The decrease has been attributable, in part, to an increased vascular superoxide anion production, which inactivates NO.…”

“…A 1-cm-long incision was made in the midscapular region, and an osmotic minipump (Alzet, model 2004) was implanted. Angiotensin II, contained in the osmotic minipumps, was dissolved in 0.15 mM NaCl containing 0.01 N acetic acid as described previously (Sarr et al, 2006). The infusion rate was 0.4 mg/kg/day.…”

“…Although an inhibition of Ang II-induced hypertension by crude extracts of red wine polyphenol was reported in rat, 26 the effect of resveratrol on Ang II-induced hypertension remains to be determined. We clearly showed that resveratrol inhibited Ang II-induced hypertension.…”

“…Indeed, it is now accepted that polyphenols could prevent many diseases such as cancers (Yang et al, 2009;Korkina et al, 2009;Khan et al, 2009;Bracke et al, 2008;Franklin and McCubrey, 2007), cardiovascular (Madeira et al, 2009;Jones et al, 2011;Kumar et al, 2008;Agouni et al, 2009;Walter et al, 2008;SchiniKerth et al, 2011;Sall Diallo et al, 2008;Sarr et al, 2006 ; and degenerative diseases (Mollnau et al, 2006;Chen et al, 2007;Berrino, 2002;Meydani, 2002;Meydani, 2001). Encouraging the consumption of fruits and vegetables is now a major public health recommendation.…”
