Red Flour Beetle (Tribolium) Germline Transformation and Insertional Mutagenesis

Abstract: INTRODUCTIONThe procedure for introducing transgenes into the Tribolium genome is similar to that for Drosophila and is of comparable efficiency. Purified transposon vector and helper plasmids are injected through the posterior pole of precellular embryos. The injected eggs are incubated under humidified conditions until they hatch. Adults resulting from injected eggs are mated inter se, and transformants are identified among their offspring with the help of visible genetic markers. The helper plasmids express… Show more

“…Starting with classic genetic screens conducted in the 1980s (Beeman et al, 1989;Sulston and Anderson, 1996), the Tribolium research community has grown, bringing a plethora of genetic tools and approaches to this species. These approaches include transgenesis (Berghammer et al, 1999(Berghammer et al, , 2009Lorenzen et al, 2003;Pavlopoulos et al, 2004), RNAimediated knockdown (Bucher et al, 2002;Posnien et al, 2009), enhancer trapping (Lorenzen et al, 2003;Trauner et al, 2009), heatshock and GAL4/UAS-mediated mis-expression (Schinko et al, 2012(Schinko et al, , 2010 and live imaging (Benton et al, 2013;Sarrazin et al, 2012;Strobl and Stelzer, 2014). Moreover, the Tribolium genome is sequenced and well annotated (Kim et al, 2010;Richards et al, 2008).…”

“…Second, Tribolium provides many tools for functional studies. It possesses a particularly strong systemic RNAi response -which was the basis for a recent genome-wide RNAi screen -and techniques for forward genetic screens, transgenesis and live imaging are available (Benton et al, 2013;Berghammer et al, 1999Berghammer et al, , 2009Koelzer et al, 2014;Sarrazin et al, 2012). Therefore, we have chosen Tribolium for unbiased genomic approaches like ChIP-seq (D. Stappert, Two novel, complementary next generation sequencing approaches to reveal the dorso-ventral gene regulatory network of Tribolium castaneum.…”

“…Again, the use of native Tribolium promoters should enhance the efficiency of these types of experiments. For details on creating transgenic lines of Tribolium, see Red Flour Beetle (Tribolium) Germline Transformation and Insertional Mutagenesis (Berghammer et al 2009). …”
