Red blood cell membrane-coated upconversion nanoparticles for pretargeted multimodality imaging of triple-negative breast cancer

Abstract: Upconversion nanoparticles (UCNPs) have been widely employed for tumor imaging using magnetic resonance imaging (MRI) and upconversion luminescence (UCL) imaging.

“…Constructs that employ RBC membranes as a substrate coating have also been shown to be stable, with many constructs showing minimal changes in size after storage of up to one week [ 42 , 43 ]. We have demonstrated that after 12 h of storage in the dark at 4 and 37 °C, fluorescence emission of nEGs loaded with ICG (ICG-nEGs) was retained, whereas there was nearly a 40% reduction in the emission for free ICG [ 44 ].…”

“…Multiple groups have investigated combining UCNPs with RBC-based delivery constructs, particularly for cancer imaging. For example, Li et al prepared NaGdF 4 :Yb, Tm UCNPs that were then coated with RBC membranes [ 42 ]. The resulting constructs showed increased tumor uptake compared to bare UCNPS, and could be used for UCL, as well as magnetic resonance imaging (MRI) and positron emission tomography (PET) imaging [ 42 ].…”

“…In a different effort, upconverting NPs (NaGdF 4 :Yb,Tm) coated with RBC membranes and an incorporated linker for click chemistry were accumulated in a tumor site in a mouse model. [18] Then, modified PET agent 18 F was injected and attached to the preaccumulated NPs via click chemistry. A PET signal from the tumor site was detectable after 30 min, indicating the feasibility of the approach.…”

“…Immune evasion and speci c tumor-targeting characteristics remain challenges to tackle to maximize the e cacy of PDT. Our previous work revealed that cancer cell membranes (CCm) might possess the desired immune evasion and homologous targeting characteristics [18,19]. Here, we designed the biomimetic oxygen delivery nanoprobe, namely cancer cell membrane-coated HSA-ICG-doped per uorotributylamine (CCm-HSA-ICG-PFTBA) for homologous targeting and hypoxia relieving at tumor sites.…”
