Recovery of Viable CD34+ Cells from Cryopreserved Haemopoietic Stem Cell Products.

Abstract: The recovery of viable CD34+ cells reinfused into patients at the time of autologous or allogeneic transplantation is clinically an important variable, which can determine graft success or failure. In this study we analyse the recovery of viable CD34+ cells /kg pre and post cryopreservation on a total of 86 autologous stem cell products from adult and paediatric patients as well as 4 cryopreserved stem cell products from allogeneic donors. CD34 enumeration was performed on all samples pre and po… Show more

“…Harvested marrow was filtered with a 200‐μm in‐line filter in the BMT laboratory. A small aliquot was taken for routine testing and counts including WCC, blood group confirmation, microbial studies and viable CD34 and CD3 cell number enumeration by flow cytometry, using a modified single platform ISHAGE gating method . Antibody panel includes: CD34‐PE (clone 8G12; Becton Dickinson, Franklin Lakes, NJ, USA), CD45‐FITC (clone 2D1; Becton Dickinson) and 7‐AAD (Becton Dickinson).…”
