Recovery of serum proteins using cellulosic affinity membrane modified by immobilization of Cu2+ ion

Abstract: SYNOPSISAn affinity membrane was prepared from a porous cellulose membrane, and adsorption and recovery of serum proteins were investigated from the viewpoint that affinity membranes are efficacious against separation and purification of biomaterials. Into the cellulose membrane, iminodiacetate (IDA) group that acts as a ligand to metal ions was introduced (Cell-IDA membrane), and then Cu2+ ion was immobilized (Cell-IDA-Cu membrane). Bovine serum albumin (BSA) and y-globulin (BrG), which are the major proteins… Show more

“…Now-a-days, polymer is the most popular material for membrane manufacture. Commercially many polymers [2] such as polyacrylonitrile [3], glycidyl methacrylate and cellulose derivatives [4] are involved to make a synthetic membrane , Previous studies have also reported chitosan as an excellent matrix of membrane compared to other polymers due to its low cost, non-toxicity and easy biodegradability [5,6]. The reason for choosing chitosan was due to its linear structure composed of polymeric 2-amino-2-deoxy-␤-d-glucose.…”

“…Cellulose acetate (CA) is a popular substrate for adsorptive membranes with hydrophilic surfaces and low non-specific protein binding. However, membranes have to be deacetylated to obtain reactive hydroxyl groups with pore size changed and loss of rigidity [16]. Among reactive groups such as hydroxyl group, amine group and sulfhydryl group, amine groups and sulfhydryl groups are much more active than hydroxyl groups and can be much more easily attacked by modifying agents (which provide functional groups) under mild conditions [17,18].…”

“…Macroporous cellulose membranes were indirectly obtained by deacetylating the cellulose acetate membranes with a methanolic KOH solution at room temperature for 6 h (73–75). These membranes are hydrophilic and pliable and exhibit little swelling at various pHs.…”
