Recovery of Purification-Associated Reduction in Antigen-Induced Histamine Release from Rat Peritoneal Mast Cells.

Abstract: The mast cell is an important target cell in the IgE-mediated allergic reaction and releases a variety of mediators upon stimulation with specific antigens.1-3) Many humoral factors and extracellular matrix proteins are reported to cause or modulate the mediator release from mast cells. [4][5][6][7][8][9][10][11][12] The peritoneal mast cells of rats, which are obtained as single cells, have been widely used for studying allergic mediator release, [13][14][15][16] and the cells are easily separated by density … Show more

“…Peritoneal exudates were collected and filtered through a 70 µm cell strainer, centrifuged at 1200 rpm for 5 minutes and re-suspended in 1 ml of sterile physiological saline. Mast cells were then purified by Percoll density gradient as previously described [32]. The purified mast cells were then cytocentrifuged at 350 x g for 5 minutes onto glass slides and were left to dry overnight before fixation in Carnoy’s fixative.…”

“…For some experiments, peritoneal mast cells (PMCs) were enriched by centrifugation using a Percoll gradient (MC purity, 60%-80%) as described. 27 Peripheral and mesenterial LNs were excised and single-cell suspensions were prepared by homogenization in PBS. …”

“…Mast cells were isolated by peritoneal lavage as previously described (Berdún et al, 2015) with some modifications (Coll et al, 2020). Peritoneal mast cells were purified by centrifugation through a discontinuous gradient of Percoll (Inagaki et al, 2001). Cells were stained with toluidine blue (0.1% (w/v), pH 1.0) and quantified using a Neubauer hemocytometer under a Nikon microscope (magnification 400×).…”
