Reconstitution of Excitable Cell Membrane Structure in Vitro

Abstract: Single stable bimolecular lipid and proteolipid membranes having the inert physical properties of cell membrane can be reconstituted in saline solution. After adsorption of appropriate molecules, these membranes become electrically excitable.

“…Phospholipids were dissolved in nheptane to a nal concentration of 20 mg/ml. BLM was formed by the procedure of Mueller et al (13) on an aperture in a cylindrical Te on chamber placed in a 12-ml glass cuvette. The working surface of BLM was 0.12 mm 2 .…”

“…However, the magnitude of the selectivity for K + over Na + was much less with PV (10-102) than it was with val (103-104) (9). For an unmodified bilayer, the I-V characteristic was linear over the range -I120 mV (16)(17)(18)(19). However, at all concentrations of PV tested, the I-V curves were nonlinear and I approached a maximum value (Im.x) independent of V when V exceeded 100 mV (Fig.…”

“…Inspired by these studies, in the early 1960's, Müller, Rudin, Tien and Wescott presented a way to prepare freestanding lipid bilayers, which are bordered by two aqueous reservoirs [MRTW62b,MRTW62a] (see figure 1.1(a)). According to previous works in history they called their membranes "Black Lipid Membranes" (BLMs).…”
