New conductive materials for tissue engineering are needed for the development of regenerative strategies for nervous, muscular, and heart tissues. Polycaprolactone (PCL) is used to obtain biocompatible and biodegradable nanofiber scaffolds by electrospinning. MXenes, a large class of biocompatible 2D nanomaterials, can make polymer scaffolds conductive and hydrophilic. However, an understanding of how their physical properties affect potential biomedical applications is still lacking. We immobilized Ti 3 C 2 T x MXene in several layers on the electrospun PCL membranes and used positron annihilation analysis combined with other techniques to elucidate the defect structure and porosity of nanofiber scaffolds. The polymer base was characterized by the presence of nanopores. The MXene surface layers had abundant vacancies at temperatures of 305− 355 K, and a voltage resonance at 8 × 10 4 Hz with the relaxation time of 6.5 × 10 6 s was found in the 20−355 K temperature interval. The appearance of a long-lived component of the positron lifetime was observed, which was dependent on the annealing temperature. The study of conductivity of the composite scaffolds in a wide temperature range, including its inductive and capacity components, showed the possibility of the use of MXene-coated PCL membranes as conductive biomaterials. The electronic structure of MXene and the defects formed in its layers were correlated with the biological properties of the scaffolds in vitro and in bacterial adhesion tests. Double and triple MXene coatings formed an appropriate environment for cell attachment and proliferation with mild antibacterial effects. A combination of structural, chemical, electrical, and biological properties of the PCL−MXene composite demonstrated its advantage over the existing conductive scaffolds for tissue engineering.

Room-Temperature Copper-Mediated Radiocyanation of Aryldiazonium Salts and Aryl Iodides via Aryl Radical Intermediates

Radiocyanation is an attractive strategy for incorporating carbon-11 into radiotracer targets, particularly given the broad scope of acyl moieties accessible from nitriles. Most existing methods for aromatic radiocyanation require elevated temperatures (Cu-mediated reactions of aryl halides or organometallics) or involve expensive and toxic palladium complexes (Pd-mediated reactions of aryl halides). The current report discloses a complementary approach that leverages the capture of aryl radical intermediates by a Cu–11CN complex to achieve rapid and mild (5 min, room temperature) radiocyanation. In a first example, aryl radicals are generated via the reaction of a CuI mediator with an aryldiazonium salt (a Sandmeyer-type reaction) followed by radiocyanation with Cu–11CN. In a second example, aryl radicals are formed from aryl iodides via visible-light photocatalysis and then captured by a Cu–11CN species to achieve aryl–11CN coupling. This approach provides access to radiocyanated products that are challenging to access using other methods (e.g., ortho-disubstituted aryl nitriles).

Meta‐signature guided investigation of miRNA candidates as potential biomarkers of oral cancer

Objectives This study aimed to experimentally validate dysregulated expression of miRNA candidates selected through updated meta‐analysis of most commonly deregulated miRNAs in oral cancer and to explore their diagnostic and prognostic potential. Materials and methods Five miRNAs (miR‐31‐3p, miR‐135b‐5p, miR‐18a‐5p, miR‐30a‐5p and miR‐139‐5p) from updated meta‐signature were selected for validation by qRT‐PCR method in 35 oral cancer clinical specimens and adjacent non‐cancerous tissue. Results Updated meta‐analysis has identified 13 most commonly deregulated miRNAs in oral cancer. Seven miRNAs were consistently up‐regulated (miR‐21‐5p, miR‐31‐3p, miR‐135b‐5p, miR‐31‐5p, miR‐424‐5p, miR‐18a‐5p and miR‐21‐3p), while five were down‐regulated (miR‐139‐5p, miR‐30a‐3p, miR‐375‐3p, miR‐376c‐3p and miR‐30a‐5p). Increased expression of miR‐31‐3p and miR‐135b‐5p, and decreased expression of miR‐139‐5p and miR‐30a‐5p were confirmed in oral cancer compared to adjacent non‐cancerous tissue. A three miRNAs combination (miR‐31‐3p, miR‐139‐5p and miR‐30a‐5p) gave the most promising diagnostic potential for discriminating oral cancer from non‐cancerous tissue (AUC: 0.780 [95% CI: 0.673–0.886], p < 0.0005, sensitivity 94.3%, specificity 51.4%). High expression of miR‐135b‐5p, miR‐18a‐5p and miR‐30a‐5p was associated with poor survival (p = 0.003, p = 0.048, p = 0.016 respectively). Conclusion miR‐31‐3p, miR‐139‐5p and miR‐30a‐5p panel was confirmed as a potential diagnostic biomarker when distinguishing oral cancer from non‐cancerous tissue. miR‐135b‐5p, miR‐18a‐5p and miR‐30a‐5p might serve as potential biomarkers of poor survival of oral cancer patients.
