Aims: The aim of this research was to characterize the metabolic pathway for maltose utilization in Enterococcus faecalis. Methods and Results: Screening a library of Enterococcus faecalis insertional mutants allowed the isolation of mutants affected in maltose utilization. Genetic analysis of the insertion loci revealed insertions in neighbour genes encoding an EII component of a phosphotransferase system (PTS) transporter (malT) and a maltose phosphorylase homologue (malP). The malP gene forms part of an operon which also includes genes encoding a phosphoglucomutase (malB), a mutarotase (aldose 1-epimerase) (malM) and a transcriptional regulator (malR). Analysis of 14 C-labelled carbohydrates uptake revealed that more than 97% of maltose enters the cells by the PTS transporter MalT. Conclusions: Both experimental data and genetic organization of the malPBMR operon strongly suggest that in Enterococcus faecalis, maltose enters using a PTS, leaving maltose-6-phosphate inside the cells which is hydrolysed by a maltose phosphate phosphorylase (MalP). Significance and Impact of the Study: This study describes a new pathway for maltose utilization in lactic acid bacteria.

The molecular organization and functional characteristics of the PAD1 replicon-encoded par stability determinant were examined. par encodes two convergently transcribed RNAS of approximately 210 and 65 nucleotides designated RNA I and RNA II, respectively. The sequence of RNA II is largely complementary to RNA I, suggesting that RNA II could regulate RNA I function as an anti-sense RNA. Results of functional studies are consistent with a role for par as a post-segregational killing system, the first to be identified in Gram-positive bacteria, with RNA I encoding the toxin and RNA II the antidote. These results include: (i) destabilization of par-containing replicons in the presence of a second complete par or the RNA II coding sequence in the same cell; (ii) par-dependent stabilization of a highly unstable vector at the expense of host-cell growth rate; and (iii) protection of cells from the toxic effects of overexpression of RNA I by RNA II supplied in trans.

Aims: To assess the inhibitory activity on Gram‐positive and Gram‐negative bacteria of several species of enterococci recovered from a natural corn silage. Methods and Results: The inhibitory activity of strains of Enterococcus faecalis (58), Enterococcus faecium (35), Enterococcus gallinarum (3) and Enterococcus casseliflavus (4) were studied employing indicator strains from various sources (clinical, food and ATCC). Enterococcus faecalis MR99, the only strain with inhibitory activity, inhibited other enterococci, Listeria spp., Staphylococcus aureus, Clostridium spp., Bacillus spp., Escherichia coli, Shigella sonnei and Shigella flexneri. The bacterium contained only one conjugative pheromone‐responsive plasmid. The partially chromatography‐purified MR99 enterocin (PPE) had a molecular weight of approx. 5000 Da and a pI of 6·2, was sensitive to proteolytic enzymes and could be extracted in benzene and butanol. It appeared stable to adjustment of pH 4·0, 5·0, 6·0, 7·0 and 8·0 and was resistant to heat. Inactivation was at 15 min at 121°C. Enterocin MR99 was bactericidal on strains of Listeria monocytogenes, Staph. aureus, and bovine mastitis agents, it was bacteriostatic on E. coli. Although enterocins MR99 and AS48 have inhibitory activity on Gram‐negative bacilli, PCR studies demonstrated a lack of relationship between them. Conclusions: The active component had a protein nature, was resistant to heat and presented a wide inhibitory spectrum. Significance and Impact of the Study: The biological properties of Ent. faecalis MR99 suggest that this strain merits further investigations so it can be applied in human and veterinary health programmes.
