Recombination-deficient mutant of Streptococcus faecalis

Abstract: An ultraviolet radiation-sensitive derivative of Streptococcus faecalis strain JH2-2 was isolated and found to be deficient in recombination, using a plasmidplasmid recombination system. The strain was sensitive to chemical agents which interact with deoxyribonucleic acid and also underwent deoxyribonucleic acid degradation after ultraviolet irradiation. Thus, the mutant has properties similar to those of recA strains of Escherichia coli. The absence of known transducing phages and the inability to transform h… Show more

“…The present study was performed using the Ent. faecalis strain JH2‐2 (Yagi and Clewell 1980) which was obtained from the parental strain JH2 (Jacob and Hobbs 1974). Enterococcus faecalis was grown at 37°C in M17 media supplemented with 0·5% glucose (GM17) or with 0·5% maltose (MM17) (Terzaghi and Sandine 1975).…”

“…coli strains JM101 (Sambrook et a/., 1989) or Sure (Stratagene Cloning Systems). RNA production and functional characterizations were carried out in E. faecalis strain OGlX (Ike et a/., 1983) or in the recombinationdeficient strain UV202 (Yagi and Clewell, 1980). Plasmids used in this study are described in Table 2.…”

“…Enterococcus faecalis JH2-SS, resistant to streptomycin [minimum inhibitory concentration (MIC): 500 lg ml )1 ] and spectinomycin (MIC: 500 lg ml )1 ) and the strains of Ent. faecalis JH2-2 and DS16 (Tomich et al 1979;Yagi and Clewell 1980) provided by Dr M Gilmore (Department of Ophthalmology, Oklahoma University, USA), were chosen for electroporation and conjugation studies and pheromone production.…”
