The aim of the investigation was to create multifunctional antitumor protein agent DARPin-mCherry-PE40 possessing both diagnostic and therapeutic properties.Materials and Methods. The experiments were carried out on human cell lines of breast adenocarcinoma SK-BR-3. Chinese hamster ovarian cell line CHO-K1 was used as a control. Genetic engineering methods, metal chelate affinity chromatography, confocal microscopy and spectrophotometric MTT test were used to perform the experiments.Results. A multifunctional targeted anticancer protein agent DARPin-mCherry-PE40 consisting of targeted HER2-specific nonimmunoglobulin module DARPin, an imaging module mCherry and cytotoxic module based on a fragment of Pseudomonas exotoxin A (PE40) has been created. Using HER2-positive cells it was shown in vitro that all three domains of the recombinant protein retained their functional qualities -high affinity for HER2 antigen, ability to fluorescence, and cytotoxicity.Conclusion. The ability of the targeted antitumor agent DARPin-mCherry-PE40 selectively stains HER2-positive cells and highly selective cytotoxicity against these cells make this targeted recombinant protein DARPin-mCherry-PE40 a perspective theranostic agent for the diagnosis and treatment of HER2-positive human tumors.

Objectives Monoclonal antibody-based of cancer therapy has been considered as one of the most successful therapeutic strategies for both haematologic malignancies and solid tumours in the last two decades. Epidermal growth factor receptor (EGFR) family signalling pathways play a key role in the regulation of cell proliferation, survival and differentiation. Hence, anti-EGFR family mAbs is one of the most promising approaches in cancer therapy. Key findings Here, recent advances in anti-EGFR mAb including approved or successfully tested in preclinical and clinical studies have been reviewed. Although we focus on monoclonal antibodies against the EGF receptor, but the mechanisms underlying the effects of EGFR-specific mAb in cancer therapy, to some extend the resistance to existing anti-EGFR therapies and some therapeutic strategies to overcome resistance such as combination of mAbs on different pathways are briefly discussed as well. Summary The EGFR family receptors, is considered as an attractive target for mAb development to inhibit their consecutive activities in tumour growth and resistance. However, due to resistance mechanisms, the combination therapies may become a good candidate for targeting EGFR family receptors.

209Направленный токсин представляют собой гибридный белок, состоящий из двух фрагментов. Первый, адресный, фрагмент способен высокоафинно связываться с онкоспецифическим рецептором на поверхности клеток. Второй фрагмент является частью белкового токсина и при попадании в клетку убивает её. Развитие идеи использования направленных токсинов продолжается уже более двух десятков лет и привело к созданию двух одобренных к применению препаратов: Ontak™ для лечения рецидивирующей кожной Т-клеточной лимфомы и препарата moxetumomab pasudotox для терапии волосатоклеточного лейкоза. Однако, несмотря на имеющиеся успехи, большинство разработок направленных токсинов окончилось неудачей. Основной причиной неудачного завершения испытаний направленных токсинов была высокая неспецифическая активность препаратов [1]. Неспецифическая активность направленных токсинов может объясняться наличием целевых рецепторов на поверхности не только опухолевых, но и здоровых клеток. Другая причина неспецифической активности может быть связана со взаимодействием с поверхностью клеток не адресной части токсина, а фрагмента, несущего цитотоксическую функцию [2]. Какие последовательности токсичного фрагмента отвечают за неспецифическое взаимодействие с клетками и возможно ли их удаление с сохранением цитотоксичности токсина, в настоящее время остаётся малоизученным вопросом. Так, было показано, что удаление транслокационного домена во фрагменте псевдомонадного токсина приводит к незначительному падению токсичности направленного токсина и многократному увеличению максимально переносимой дозы у мышей [3]. Авторы этой работы предположили, что данный эффект связан с уменьшением гепатотоксичности модифицированного варианта токсина у мышей. Стоит отметить, что сравнение гепатотоксичности токсинов на клетках человека в этой работе проведено не было. Ранее нами были разработаны направленные токсины DARPin-PE40 [4] и DARPin-LoPE [5], селективно уничтожающие клетки с повышенным содержанием онкоспецифического рецептора ERBB2. Этот рецептор часто используется при разработке различных направленных противоопухолевых агентов [6]. Различие между разработанными нами токсинами заключается в использовании в токсине DARPin-LoPE низкоимунногенного варианта фрагмента псевдомонадного токсина с удалённым транслокационным доменом. Целью на-УДК 577.29 Поступило 15.08.2019 г.Направленные токсины являются перспективными противораковыми агентами, позволяющими селективно уничтожать раковые клетки за счёт повышенного содержания на их поверхности онкоспецифических маркёров. Применение таких противораковых токсинов в медицине сдерживается в основном их высокой неспецифической токсичностью, в особенности гепатотоксичностью. В экспериментах на клетках человека мы показали, что удаление транслокационного домена токсина DARPin-PE40 приводит к уменьшению гепатотоксичности и неспецифической активности. Такой же эффект наблюдается и при инактивации в молекуле DARPin-PE40 сайта расщепления фурином. Одновременное удаление как транслокационного домена, так и сайта расщепления фурином показал...
