Recombinant Respiratory Syncytial Virus with the G and F Genes Shifted to the Promoter-Proximal Positions

Abstract: The genome of human respiratory syncytial virus (RSV) encodes 10 mRNAs and 11 proteins in the order 3-NS1-NS2-N-P-M-SH-G-F-M2-1/M2-2-L-5. The G and F glycoproteins are the major RSV neutralization and protective antigens. It seems likely that a high level of expression of G and F would be desirable for a live RSV vaccine. For mononegaviruses, the gene order is a major factor controlling the level of mRNA and protein expression due to the polar gradient of sequential transcription. In order to increase the expr… Show more

“…Wild-type RSV was further purified by centrifugation through a sucrose gradient to eliminate cytokine contaminants (58). Virus concentrations were determined by plaque assay (28). RSV was inactivated with UV light (UV-RSV) in a Stratalinker UV cross-linker for 10 min (0.12 J/cm 2 ; Stratagene, La Jolla, CA).…”

“…One strategy to increase the level of expression of G and F protective antigens is to move their genes so as to be immediately downstream of the viral promoter. This was done for F and G individually and in combination, and resulted in two-to threefold increase in their synthesis without changing the virulence of the virus (41). Although this is a modest effect, it would be an important improvement if it were reflected in a comparable increase in immunogenicity in young infants.…”

“…Here we have explored the distinctive organization of the arenavirus genome as an unique opportunity for the rational design of live attenuated vaccines based on rearrangement of the viral genes to disrupt normal viral gene expression and, therefore, viral fitness. The rationale for this approach is further supported by current efforts aimed at evaluating reorganization of the viral genome as a novel strategy for the generation of live attenuated vaccine candidates for a variety of DNA and RNA viruses (37)(38)(39)(40)(41)(42)(43)(44)(45).…”
