Recombinant parvovirus-like particles as an antigen carrier: A novel nonreplicative exogenous antigen to elicit protective antiviral cytotoxic T cells

Abstract: To develop a strategy that promotes efficient antiviral immunity, hybrid virus-like particles (VLP) were prepared by self-assembly of the modified porcine parvovirus VP2 capsid protein carrying a CD8 ؉ T cell epitope from the lymphocytic choriomeningitis virus nucleoprotein. Immunization of mice with these hybrid pseudoparticles, without adjuvant, induced strong cytotoxic T lymphocyte (CTL) responses against both peptide-coated-or virus-infected-target cells. This CD8 ؉ class I-restricted cytotoxic activity pe… Show more

“…Wild-type (WT) PPV VLPs and chimeric PPVOVA VLPs carrying the OVA 257-264 epitope were produced using a BV expression system (13). VLPs were precipitated with 20% ammonium sulfate and dialyzed.…”

“…This pathway may be exploited for the development of nonreplicating, safe vaccines that nevertheless induce strong CTL responses. A variety of exogenous Ags have been shown to induce effective CTL responses in vivo, including Ags associated with apoptotic cells (10,11), cell debris (7,12,13), heat shock proteins (14), Sephadex beads (5), and virus-like particles (VLPs) (6,(15)(16)(17).…”

“…VLPs have also been proven to be potent CTL inducers compared with other vectors (21)(22)(23)(24). A study of PPV-VLPs immunization showed a safe and efficient strategy to induce CTLs and stimulate antiviral immunity in a CD4-independent manner (22).…”
