Recombinant Lipase Engineered with Amphipathic and Coiled-Coil Peptides

Abstract: Lipases have been utilized industrially to produce biodiesel, oleochemicals, and pharmaceuticals. Many efforts such as metagenomics, directed evolution and enzyme immobilization have been devoted to enhance the lipase activity. Here, we designed a recombinant lipase, NKC-M37-MAT, that was generated by incorporating an N-terminal amphipathic peptide (NKC) and a C-terminal coiled-coil peptide (MAT) into Photobacterium lipolyticum M37 lipase. The hydrophobic face of NKC improve the accessibility (K m ), and cataly… Show more

“…26 Similarly, a recombinant enzyme containing a c-terminal coiled-coil peptide was reported to have strong activity and high thermochemical stability compared with the wild-type enzyme. 27 Moreover, a helix-stabilized antibody fragment (hsFv) obtained by fusing a coiled coil to the Fv fragment of an antibody possessed similar expression, stability, and oligomerization properties to other Fv constructs. 28 This substitution may protect FcfVEGF-Trap from Fc region-related elimination in addition to stabilizing the FcfVEGF-Trap structure, and may eventually contribute to longer residence time.…”

“…NKC-TliA and CTP-TliA are derivatives of TliA. NKC is an antibiotic polypeptide developed in a previous study (40), and CTP is a cytoplasmic transduction peptide that was developed as a cellular import tag in a previous study (14). We have synthesized genes for these two, with codons optimized for P. fluorescens expression.…”

“…To overcome these problems, enzymes have been immobilized onto a variety of supports, including bio-related nanoarchitectures, metal-organic frameworks and hybrids, polymers, and nanomaterials 17 23 . Additionally, several immobilization strategies have been developed to increase the stability and reusability of enzymes 6 , 24 . After achieving an efficient enzyme immobilization, researchers have sought to improve the various properties of the enzymes and nanoparticles to develop an ideal biocatalyst 8 , 25 .…”
