Recombinant Human CXC-Chemokine Receptor-4 in Melanophores Are Linked to GiProtein: Seven Transmembrane Coreceptors for Human Immunodeficiency Virus Entry into Cells

Abstract: This article describes the transient expression of the CXC chemokine receptor-4 in Xenopus laevis melanophores and the resulting functional assay for the endogenous ligand for this receptor stromal cell-derived factor (SDF)-1␣. Specifically, it will be shown that SDF-1␣ produces increased light transmittance in transfected cells that is consistent with the activation of G i protein. This stimulus pathway is further implicated by the abolition of this response after pretreatment of the cells with pertussis toxi… Show more

“…Our results showed that C5a-induced response was significantly attenuated by PTX [28,29]. PLC activation is one of the earliest events that follow chemotactic agent -receptor interaction in a variety of cell type cell membrane.…”

“…CXCR4, like most other chemokine receptors, signals through G α i proteins in response to SDF-1 α stimulation (4850). G α i activation results in a variety of signaling events, including the inhibition of cAMP accumulation (49).…”

“…Stromal cell-derived factor-1 (SDF-1) is a chemokine that interacts specifically with CXCR4. When SDF-1 binds to CXCR4, the complex activates pertussis toxin-sensitive Ga I protein-mediated signaling (10). The interaction between SDF-1 and CXCR4 has been shown to direct cells to organ sites with high levels of SDF-1 expression, suggesting that this interaction plays a key role in the chemotaxis and homing of metastatic cells.…”
