Recombinant Bovine Adenovirus-3 Co-Expressing Bovine Respiratory Syncytial Virus Glycoprotein G and Truncated Glycoprotein gD of Bovine Herpesvirus-1 Induce Immune Responses in Cotton Rats

Abstract: One of the impediments in the development of safe and cost effective vaccines for veterinary use has been the availability of appropriate delivery vehicle. We have chosen to develop and use bovine adenovirus (BAdV)-3 as vaccine delivery vector in cattle. Here, we describe the construction of recombinant E3 deleted BAdV-3 vectors expressing single vaccine antigen (BAV360; bovine respiratory syncytial virus G) or two vaccine antigens (BAV851; bovine herpesvirus-1gDt and bovine respiratory syncytial virus G). Rec… Show more

“…Ten pools of five randomly selected BRD samples were assembled using 32 l from each sample. The resulting 160-l pools were passed through a 200-nm filter, and the resulting filtrates were incubated for 1.5 h in a cocktail of DNase and RNase enzymes (53). Nucleic acids were then extracted using the QIAquick viral RNA column purification system according to the manufacturer's instructions.…”

“…Moreover, two intranasal immunizations, four weeks apart of calves with single recombinant BAdV-3 expressing BHV-1 gDt and BRSV gG induced significant neutralizing antibodies against BHV-1 and BRSV in cotton rats [72]. Thus, the construction of single recombinant BAdV-3 with multiple vaccine antigens is feasible to induce virus specific immunity for protection from multiple pathogens in calves [72].…”

“…Since veterinary vaccines have to be safe and cost effective, the development of a recombinant BAdV-3 vectored vaccine providing immunity to more than one pathogen is desirable. Recently, we demonstrated that single recombinant BAdV-3 expressing vaccine antigens from two respiratory pathogens (gDt of BHV-1 and gG of bovine respiratory syncytial virus [BRSV]) of cattle could be isolated [72]. Moreover, two intranasal immunizations, four weeks apart of calves with single recombinant BAdV-3 expressing BHV-1 gDt and BRSV gG induced significant neutralizing antibodies against BHV-1 and BRSV in cotton rats [72].…”

“…129 A BAdV-3 vector (BAV851) coexpressing bovine respiratory syncytial virus G and BHV-1gDt proteins was generated, and vaccination of cotton rats with this BAV851 induced strong antigen-specific immune responses. 130 In order to evaluate whether a BAdV vector can effectively elude high levels of preexisting HAdV-5 vector immunity, naïve and HAdV-5-primed mice were immunized with BAd-H5HA (a BAdV vector expressing HA of a H5N1 influenza virus). 131 Even in the presence of very high levels of HAdV-5-specific neutralizing antibody titer (2133 ± 660), no reductions in HA-specific humoral and CMI responses were observed in mice immunized with BAd-H5HA.…”
