Recognition of specific DNA sequences by mitomycin C for alkylation

Abstract: Synthetic oligodeoxyribonucleotides were reacted with mitomycin C (MC) under conditions which restricted MC to monofunctional alkylating activity. The yields of monofunctional alkylation of oligonucleotides with variable sequence were determined by enzymatic digestion of the reaction mixture to unreacted nucleosides and the product of alkylation, a MC-deoxyguanosine adduct (2), followed by quantitative analysis by HPLC. The relative yields of 2 reflected relative monoalkylation reactivities. They were compared… Show more

“…Exposure to MMC results in alkylation and interstrand cross-links (20,26), which are mainly repaired by NER in E. coli (52). The M. smegmatis NER mutants differed in their sensitivities to MMC.…”

“…In contrast, MRD appears to confer resistance by binding MC as a complex that prevents drug activation. The formation of MC-DNA adducts is highly efficient and selective for guanine nucleosides in the specific sequence 5Ј-CpG-3Ј (27,28). On the basis of an average GϩC content of 70%, there may be as many as 10 6 target sites within the genome of a typical streptomycete.…”

“…These results demonstrate conclusively a strong correlation between duplex formation and the extent of alkylation with FR66979, analogous to that observed with MC, suggesting that monoalkylation reactions are strongly favored in duplex DNA structures. 9 …”

“…7 More is known about the interstrand cross-linking reactions of 1±3 than about their abilities to either monoalkylate or form intrastrand cross-links in DNA. In contrast, MC has been shown to form both intrastrand cross-links (at d(GG)) 8 and monoadducts (at dG) 9 in DNA through N2 of dG. Preliminary evidence for the formation of monoadducts between FR66979 and DNA was ®rst obtained in a study of DNA interstrand cross-linking by FR66979.…”
