Prostate cancer represents a major contributor to cancer mortality, but the majority of men with prostate cancer will die of other causes. Thus, a challenge is identifying potentially lethal disease at diagnosis. Conflicting results have been reported when investigating the relationship between infiltration of lymphocytes and survival in prostate cancer. One of the mechanisms suggested is the recruitment of regulatory T cells (T regs ), a subpopulation of T cells that have a role in promoting tumor growth. T regs counteract tumor rejection through suppressive functions on the anti-immune response but their prognostic significance is still unknown. We report here the results of a conducted case-control study nested in a cohort of men treated with transurethral resection of the prostate and diagnosed incidentally with prostate cancer. Cases are men who died of prostate cancer (n ¼ 261) and controls are men who survived 410 years after their diagnosis (n ¼ 474). Infiltration of both T helper and T cytotoxic cells was frequently observed and the majority of the T regs were CD4 þ . T helper or T cytotoxic cells were not associated with lethal prostate cancer. However, we found a nearly twofold increased risk of lethal prostate cancer when comparing the highest with the lowest quartile of CD4 þ T reg cells (95% confidence interval: 1.3-2.9). Our conclusion is that men with greater numbers of CD4 þ T regs in their prostate tumor environment have an increased risk of dying of prostate cancer. Identification of CD4 þ T regs in tumor tissue may predict clinically relevant disease at time of diagnosis independently of other clinical factors.

MHC class I molecules expressed on cell surfaces are composed of H chain, β2-microglobulin and any of a vast array of peptides. The role of peptide in the recognition of HLA class I by serum HLA Abs is unknown. In this study, the solid-phase assay of a series (n = 11) of HLA-A2-reactive, pregnancy-induced, human mAbs on a panel (n = 12) of recombinant monomeric HLA-A2 molecules, each containing a single peptide, revealed peptide selectivity of the mAbs. The flow cytometry membrane staining intensities on the HLA-A2-transduced cell line K562, caused by these mAbs, correlated with the number of monomer species detected by the mAbs. Flow cytometry staining on HLA-A2-bearing cell lines of a variety of lineages was indicative of tissue selectivity of these HLA-A2 mAbs. This tissue selectivity suggests that the deleterious effect on allografts is confined to alloantibodies recognizing only HLA class I loaded with peptides that are derived from tissue-specific and household proteins. Since Abs that are only reactive with HLA loaded with irrelevant peptides are expected to be harmless toward allografts, the practice of HLA Ab determination on lymphocyte-derived HLA deserves reconsideration.

Gastric cancer is the most common malignancy in the world. Often diagnosed at advanced and unresectable stage, there is no curative treatment available for gastric cancer. Immunotherapy might be a promising alternative option. The key challenge in gastric cancer diagnosis and immunotherapy is to find an efficient target that not only provide early indication of the disease but also can upregulate the anti-tumor immune response in the cancer patients. Tumor-associated antigens(TAAs) expressed in tumor cells can elicit the immune response in autologous host and are related to changes during the disease process may fulfill this demanding. Now, Thousands of antigens have been discovered by a powerful method called SEREX(serological analysis of tumor antigen by recombinant cDNA expression libraries) and some of SEREX-defined antigens have been evaluated in clinical trails as vaccines for cancer immunotherapy. This review will provide information on the application of SEREX method in gastric cancer research and knowledge learned from these studies to develop antigen-specific diagnosis and therapy for gastric cancer.
