Recipient Glycemic Micro-environments Govern Therapeutic Effects of Mesenchymal Stem Cell Infusion on Osteopenia

Abstract: Therapeutic effects of mesenchymal stem cell (MSC) infusion have been revealed in various human disorders, but impacts of diseased micro-environments are only beginning to be noticed. Donor diabetic hyperglycemia is reported to impair therapeutic efficacy of stem cells. However, whether recipient diabetic condition also affects MSC-mediated therapy is unknown. We and others have previously shown that MSC infusion could cure osteopenia, particularly in ovariectomized (OVX) mice. Here, we discovered impaired MSC… Show more

“…1c, d ). Furthermore, the loss of 661 W cell viability could be attributed to provoked cell death by H 2 O 2 and MNU, as shown by increased positively stained proportion of propidium iodide (PI) for dead cells 25 , 26 (Supplementary Fig. 1A) .…”

“…Briefly, after treatment by H 2 O 2 or MNU, or transfected with lentiviral vectors, cells were incubated with 20-μl 5 mg/ml MTT (MP Biomedicals, USA) for 4 h. The precipitates were extracted with 180-μl DMSO and the absorbance was measured at the optical density (OD) of 490 nm as the viability index. Cell death was determined by PI staining, as previously reported 25 , 26 , according to manufacturer’s instruction (Sigma-Aldrich, USA).…”
