Receptor protein tyrosine kinase EphB4 is up-regulated in colon cancer

Abstract: Background: We have used commercially available cDNA arrays to identify EphB4 as a gene that is up-regulated in colon cancer tissue when compared with matched normal tissue from the same patient.

“…The Ephrin type B receptor 4 and its ligand Ephrin B2 appear to provide critical guidance cues at points of cell-cell contact in cardiovascular development. In adult settings of neo-angiogenesis, such as in tumors, it appears that the endothelium of a subset of new vessels strongly expresses the Ephrin B2 ligand (24), whereas the tumor cells express both the ligand and the receptor (25,26). Interestingly, it has been demonstrated that the Ephrin type B receptor 4 is up-regulated in colon tumors when compared with normal colon tissue from the same patient (26).…”

As such, the Ephrin type B receptor 4 may provide angiogenic signaling for the recruitment of new blood vessels to support tumor growth.…”

“…EphB4 is normally expressed on venous endothelial cells, while its exclusive ligand, EphrinB2, is expressed on arterial endothelial cells as a transmembrane protein as well. Aberrant expression of EphB4 has been observed in several tumor cell types, although its role in tumor cell biology remains to be fully elucidated (Andres et al, 1995;Berclaz et al, 1996Berclaz et al, , 2003Stephenson et al, 2001;Takai et al, 2001). EphB4 activation leads to a number of downstream effects, which influence cell attachment, migration and interaction with ligand-expressing cells (Adams et al, 1999;Oates et al, 1999;O'Leary and Wilkinson, 1999;Steinle et al, 2002).…”

“…They also play a role in angiogenesis (Favre et al, 2003;Sullivan and Bicknell, 2003). In recent years, some of the Eph genes have been found to be overexpressed in human tumors, including neuroblastoma (Tang et al, 1999), lung carcinoma (Tang et al, 1999), gastric cancer (Kiyokawa et al, 1994;Kataoka et al, 2002), esophageal cancer (Miyazaki et al, 2003), breast cancer (Wu et al, 2004), and colorectal cancer (Stephenson et al, 2001;Liu et al, 2002Liu et al, , 2004Kataoka et al, 2004). Among the Eph family genes, relatively less attention has been directed toward EphA7 in human tumors, and the potential role of EphA7 in human oncology has not been addressed.…”
