Recent trends in the development of vitamin B12derivatives for medicinal applications

Abstract: This Feature Article highlights recent developments in the field of vitamin B12 derivatives for medicinal applications. The following topics are emphasized: (1) the development of aquacorrinoids for cyanide detection and detoxification, (2) the use of vitamin B12 conjugates and (3) antivitamins B12 for therapy and diagnosis, and (4) the design of corrinoids as activators of soluble guanylyl cyclase (sGC).

“…This approach seemed to be reasonable because small variations in the coordination sphere of rhenium complexes have profound consequences on the electrochemistry, water stability and CO releasing properties of the dicarbonyl core. 17 B 12 appeared to be attractive as ligand for the rhenium-based CORM entity mainly because of two reasons: (a) its cellular uptake properties can be exploited to deliver therapeutic agents specifically at disease sites; [18][19][20] (b) the electronic properties at the cobalt center can be selectively modified by introducing structural modifications at the corrin-p-system. [21][22][23][24] Having the general design of the B 12 -ReCORMs derivatives in mind (Scheme 1), we synthesized and studied first a series of six cobalamins for coordinating them later to 1 (Scheme 2A).…”

“…Soc. 137 (2015) -Optimized procedures are developed for the fluorination of 2-substituted ring systems and for di-or monofluorination of unsubstituted chains. In all cases, the NEt 3 ·3HF/Ph-I(O-Ac) 2 combination works with optimal results and the protocol is compatible with a gram-scale access.…”

“…-(SOBHANI*, S.; ZERAATKAR, Z.; ZARIFI, F.; New J. Chem. 39 (2015) (bpd) ligand is prepared and anchored on the surface of magnetic nanoparticles (MNP). The ligand and the surface functionalized nanoparticles of type MNP@bpd are fully characterized and are proved to be excellent catalysts after in situ addition of the rhodium metal precursor for the hydroformylation reaction of styrene and its derivatives.…”
