Recent Trends in Research with Human Pluripotent Stem Cells: Impact of Research and Use of Cell Lines in Experimental Research and Clinical Trials

Abstract: SummaryThe human pluripotent stem cell (hPSC) research landscape is rapidly evolving. To assess possible novel trends in hPSC usage, we analyzed experimental hPSC research published from 2014 to 2016 and compared our data with those of earlier periods. The number of papers describing experimental work involving hPSCs increased further with clear differences in the scientific impact of publications from different countries. Our results confirm the leading position of US-based hPSC research, although to a lesser… Show more

“…Inevitably, differences between donor individuals have been found to affect most iPSC cellular traits, from DNA methylation, mRNA and protein abundance to pluripotency, differentiation and cell morphology (Kilpinen et al, 2017). High variability in differentiation potential and genetic stability between iPSC lines remain subjects of intense research (Guhr et al, 2018). Moreover, even after controlling for genotype, substantial experimental heterogeneity remains.…”

“…So far, comparison of MSCs and iMSCs primarily focused on the similarities in cell surface marker expression, cellular morphology, differentiation potential, and immunomodulatory function-and these aspects still deserve further elucidation. In fact, the first clinical trials with iMSCs are already ongoing for steroid-refractory graft versus host disease [12,13]. With regard to the high demands of cell safety in therapeutic application, it is therefore crucial to better understand the clonal dynamics and the heterogeneity within iMSCs.…”

“…The hydrogel's 3D microenvironment supports an efficient differentiation of cells to desired lineages, and encourages physiological processes that are necessary for tissue assembly, development, and maturation. Although iPSCs are considered a promising strategy for obtaining an autologous source of cardiac and neuronal cells, there are still safety issues, such as their genomic instabilities and their ability to induce tumor formation, that must be addressed before their use in the clinic . We believe that the reported concept, where the matrix and the cells are taken from the omentum of the patient paves the way for the use of personalized implants with reduced immune response, therefore, may provide better conditions for engraftment.…”
