“…We have described recombinant sources of several S related polypeptides from two common expression systems, recombinant baculoviruses and E.coli and used the proteins expressed for the analysis of seroconversion and for epitope mapping. The particular properties of the insect cell system, yield, robustness and the ability to perform at scale are discussed elsewhere [23], similarly the use of the T7 system in E.coli [24]. Using a set of overlapping S fragments we demonstrated epitope mapping of monoclonal antibodies 3G9 and CR3022 to residues 300-400 of S. Interestingly, neither antibody reacted with the RBD itself in the western blot format used despite it encompassing the residues recognised.…”

“…In particular, the identification of two main groups of applications for engineered baculoviruses is possible: (a) systems for the expression of recombinant proteins within eukaryotic contexts and (b) the use of recombinant virions in mammals for different goals. The former are generally called Baculovirus Expression Vector Systems or simply BEVS (O'Reilly et al 1994;Possee et al 2020) and comprise platforms composed of vectors for molecular cloning that enable modifications of the AcMNPV genome (many being based on bacmids) and susceptible insect cell lines plus their culture media or a whole insect as a biofactory (Martínez-Solís et al 2019). The latter group contains engineered baculoviruses applied to mammals and is recognized as BacMam technologies (Airenne et al 2013;Mansouri and Berger 2018;Ono et al 2018).…”

“…These developments have led to different regulatory considerations because in some instances baculoviruses can be only a contaminant, while in others they are the main ingredient of the products. In contrast to the use of this technology in the production of bioinsecticides (where OBs of many viral species and production systems are used in larvae), genomeengineered BVs of AcMNPV multiplied on the Sf9 or Sf21 insect cell lines, among others, are generally the bases of the BEVS and BacMam technologies (Kwang et al 2016;Possee et al 2020).…”

“…Some of these viruses are used as bioinsecticides for the control of agricultural pests [7][8][9]. Additionally, these viruses are used as foreign protein expression systems in vitro or in vivo and as delivery systems for therapeutic genes in mammalian cells [10,11]. Baculoviruses have been isolated from different orders of insects (Lepidoptera, Hymenoptera and Diptera) and have two infective morphologies: budded virus (BV) and occlusion derived virus (ODV) [12] (Figure 1).…”
