Simple, accurate and reproducible UV spectrophotometric method was established for the assay of Fourosemide based on the formation of Schiff 's bases coupling with aromatic aldehydes such as benzaldehyde, salicylaldehyde, 2-methoxy benzaldehyde, anisaldehyde, vanilline, 2,3,4-chlorobenzaldehyde, 4-nitro benzaldehyde and dimethylaminobenzaldehyde in presence of sulphuric acid, The optical characteristics such as Beer's law limits, molar absorptivity and Sandell's sensitivity for the methods are given. Regression analysis using the method of least squares was made to evaluate the slope (b), intercept (a) and correlation coefficient (r) and standard error of estimation (SE) for each aldehyde. Determination of furosemide in bulk form and in pharmaceutical formulations were also incorporated.

Development and Validation of Furosemide Quantification Method By 1H NMR

Abstract:Furosemide is a potent diuretic used for treatment of edema, hypertension and renal disease, among others. In this work, a simple and accurate method using 1 H NMR has been developed for quantification of furosemide in pharmaceutical formulations. All evaluated parameters (selectivity, linearity, accuracy, precision, limit of detection and limit of quantification) showed satisfactory results. Five pharmaceutical formulations (tablets) were analyzed by the developed method and the method described in the Brazilian Pharmacopoeia. Results show no significant differences between them. Thus, furosemide quantification by NMR technique proved to be a promising method as an alternative for assessing the quality of medicines.Keywords: NMR; quantification; furosemide; pharmaceutical formulations. ResumoFurosemida é um potente diurético usado para o tratamento de edema, hipertensão, doenças renais, entre outras. Neste trabalho, um método simples e acurado utilizando RMN de 1 H foi desenvolvido para quantificação de furosemida em formulações farmacêuticas. Todos os parâmetros avaliados (seletividade, linearidade, precisão, exatidão, limite de detecção e limite de quantificação) apresentaram resultados satisfatórios. Cinco formulações farmacêuticas (em comprimidos) foram analisadas pelo método desenvolvido e pelo método descrito na Farmacopeia Brasileira. Os resultados não apresentaram diferenças significativas entre eles. Assim, a quantificação furosemida pela técnica de RMN provou ser um método promissor como uma alternativa para a avaliação da qualidade dos medicamentos.Palavras-chave: RMN; quantificação; furosemida; formulações farmacêuticas.

A simple, sensitive, and specific method for furosemide (FUR) analysis by reverse-phase-HPLC was developed using a Spherisorb C18 ODS 2 column. A chromatographic analysis was carried out using a mobile phase consisting of acetonitrile and 10 mM potassium phosphate buffer solution: 70 : 30 (v/v) at pH 3.85, at a flow rate of 1 mL·min−1. The UV-detection method was carried out at 233 nm at room temperature. Validation parameters including limit of detection (LOD), limit of quantitation (LOQ), linearity range, precision, accuracy, robustness, and specificity were investigated. Results indicated that the calibration curve was linear (r 2 = 0.9997) in the range of 5.2 to 25,000 ng·mL−1, with ε value equal to 3.74 × 104 L·M−1 ·cm−1. The LOD and LOQ were found to be 5.2 and 15.8 ng·mL−1, respectively. The developed method was found to be accurate (RSD less than 2%), precise, and specific with an intraday and interday RSD range of 1.233–1.509 and 1.615 to 1.963%. The stability of native FUR has also been performed in simulated perilymph and endolymph media (with respective potency in each medium of 99.8 ± 2.3% and 96.68 ± 0.7%, n = 3) after 6 hours. This method may be routinely used for the quantitative analysis of FUR from nanocarriers, USP tablets and release media related to hearing research
