QuEChERS LC–MS/MS Screening Method for Mycotoxin Detection in Cereal Products and Spices

We developed and validated a screening method for mycotoxin analysis in cereal products and spices. Ultra-high-performance liquid chromatography coupled with tandem mass spectrometry (UHPLC–MS/MS) was used for the analysis. Dispersive solid-phase extractions (d-SPEs) were used for the extraction of samples. Ochratoxin A (OTA), zearalenone (ZEA), aflatoxins (AFLA; AFB1, AFB2, AFG1, AFG2), deoxynivalenol (DON), fumonisin (FUMO; FB1, FB2, FB3), T2, and HT2 were validated in maize. AFLA and DON were validated in black pepper. The method satisfies the requirements of Commission Regulation (EC) no. 401/2006 and (EC) no. 1881/2006. The screening target concentration (STC) was under maximum permitted levels (MLs) for all mycotoxins validated. The method’s performance was assessed by two different proficiencies and tested with 100 real samples.

A fast and sensitive multi-residue analytical method was developed for the determination of organophosphorus pesticide residues in Panax notoginseng and Aucklandia lappa decne by comparing an ultrasonic extraction and homogenate extraction approach coupled with automated solid-phase extraction system and gas chromatography-mass spectrometry (GC/MS). Various factors affecting the homogenate extraction and purification efficiency were investigated in detail. The optimum conditions for homogenate extraction of organophosphorus pesticide residues from Panax notoginseng and Aucklandia lappa decne were 60 mL dichloromethane, 10000 r/min of extracting revolution speed and 2 min of extracting time. The extracts were cleaned up by automated solid-phase extraction system and determined by GC/MS. Under the optimum conditions, the results of the homogenate extraction approach were compared with those of the ultrasonic extraction, and the homogenate extraction approach was used to extract the samples. The average recoveries of the method by a homogenate extraction approach coupled with automated solid-phase extraction system and GC/MS ranged from 72.56% to 96.70%, and RSDs were 0.05~0.14%, which allowed the determination of 16 organophosphorus pesticide residues in Panax notoginseng and Aucklandia lappa decne. The results indicated that the proposed method can apply for the determination of organophosphate pesticides residues in Panax notoginseng and Aucklandia lappa decne with high accuracy and precision.

In this study, a specific and sensitive method of liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) was developed for the determination of penconazole enantiomers in rat plasma. The enantioseparation was achieved on a Chiralpak IC column by using acetonitrile/water (80:20, v/v) as the mobile phase. Penconazole enantiomers and internal standard l-lansoprazole (IS) were detected in multiple reaction monitoring (MRM) mode with positive electrospray ionization source. The method was validated over the concentration range of 2.5–250.0 ng mL−1 for penconazole enantiomers. Good linearity was obtained for both enantiomers with correlation coefficients (R) greater than 0.995. The relative error was well within the admissible range of −1.1–3.2%, and relative standard deviation was less than 6.0%. After validation, the established method was successfully applied to a stereoselective toxicokinetic study in female and male rats after oral administration of 50 mg kg−1 racemic penconazole. This is the first experiment regarding the stereospecific toxicokinetic study of penconazole and the bioanalytical approach for its quantitation in vivo.
