Recent Advances of Thiamine in Organic Synthesis

Abstract: Thiamine (VB1) has been proved to be a powerful biocatalyst for various organic transformations. VB1 offers several unique advantages, such as economical, non‐toxic, stable and water soluble. Excellent outcomes have been achieved in synthetic applications using VB1 as a catalyst. This review summarizes recent developments in the field of VB1 biocatalysis, allowing the construction of various biologically active heterocyclic scaffolds.

“…15 However, VB1 has been used as an organocatalyst for a number of organic transformations. 16 We have chosen to examine vitamin B1 (also known as thiamin, 1) as the NSHC ligand precursor because VB1, containing substituted pyrimidine connected through a CH 2 linker to the thiazolium ring, is cheap, readily available, degradable, and water soluble. It is an essential micronutrient for humans, found in food and commercially synthesized to be a dietary supplement or medication.…”
