Recent Advances in Signaling Pathways Comprehension as Carcinogenesis Triggers in Basal Cell Carcinoma

Abstract: Basal cell carcinoma (BCC) is the most common malignant skin tumor. BCC displays a different behavior compared with other neoplasms, has a slow evolution, and metastasizes very rarely, but sometimes it causes an important local destruction. Chronic ultraviolet exposure along with genetic factors are the most important risk factors involved in BCC development. Mutations in the PTCH1 gene are associated with Gorlin syndrome, an autosomal dominant disorder characterized by the occurrence of multiple BCCs, but are… Show more

“…BCC displays a different behavior compared with other neoplasms; it has a slow evolution and metastasizes very rarely, but sometimes causes significant local damage. Chronic ultraviolet exposure, along with genetic factors, is the most important risk factor for the development of BCC [ 7 ]. Whole-exome sequencing to characterize the mutational landscape of sporadic BCCs has revealed that only Patched 1 ( PTCH1 ) is associated with a significant functional mutation burden.…”

“…EGFR overexpression has been identified in various tumors, being involved in the modulation of various signaling pathways responsible for cell proliferation, invasion and metastasis. In BCC, the crosstalk between the EGFR and Hedgehog pathways induces the activation of RAS/MEK/ERK and JUN/AP-1 signaling [ 162 ].…”

“…Basal cell carcinoma (BCC) is the most common cutaneous malignancy [1]. The molecular pathogenesis of BCC is associated with the hedgehog signaling pathway and mutations in the patched homologue 1 (PCTH-1) transmembrane tumor-suppressing protein [2][3][4]. Several potential risk factors influence the development of BCC including exposure to ultraviolet radiation, genetic predisposition, genodermatoses, immunosuppression, and trauma [5].…”
