The purpose of this study was to explore the photodynamic and photothermal effects of the supramolecular material Purp@COP and to test the anti-cancer effect on HepG2 cells in vitro.Materials and methodsPurp@COP is a covalent organic polymer (COP) with robust tailoring heteroatom incorporation, plentiful pore structure, and multiple functions similar to the metal–organic framework (MOF). Hepatocellular carcinoma cell line HepG2 was cultured with Purp@COP for 24 h and treated with near-infrared 808-nm laser 1 W/cm2 for 10 min. Cell Counting Kit-8 (CCK-8) assay, colony formation assay, live–dead cell fluorescence staining, and Annexin V/propidium iodide (PI) staining flow cytometry were performed to detect the viability, proliferation, and apoptosis of the HepG2 cells.ResultsThe supramolecular material Purp@COP exhibited significant photothermal performance under near-infrared 808-nm laser irradiation in vitro. With the treatment of Purp@COP and near-infrared 808-nm laser irradiation on HepG2 cells, cell viability and colony formation capacity were decreased, and the number and proportion of apoptotic cells were increased.ConclusionsThe supramolecular material Purp@COP has both photothermal and photodynamic effects and can significantly induce cancer cell death and inhibit the proliferation of cancer cells in vitro.

Spherical gold nanoparticles (GNPs), whose unique properties regarding biomedical applications were broadly investigated, are an object of interest as nanocarriers in drug targeted delivery systems (DTDSs). The possibility of surface functionalization, especially in enabling longer half-life in the bloodstream and enhancing cellular uptake, provides an opportunity to overcome the limitations of popular anticancer drugs (such as cisplatin) that cause severe side effects due to their nonselective transportation. Herein, we present investigations of gold nanoparticle–cisplatin systems formation (regarding reaction kinetics and equilibrium) in which it was proved that the formation efficiency and stability strongly depend on the nanoparticle surface functionalization. In this study, the capillary electrophoresis hyphenated with inductively coupled plasma tandem mass spectrometry (CE-ICP-MS/MS) was used for the first time to monitor gold–drug nanoconjugates formation. The research included optimizing CE separation conditions and determining reaction kinetics using the CE-ICP-MS/MS developed method. To characterize nanocarriers and portray changes in their physicochemical properties induced by the surface’s processes, additional hydrodynamic size and ζ-potential by dynamic light scattering (DLS) measurements were carried out. The examinations of three types of functionalized GNPs (GNP-PEG-COOH, GNP-PEG-OCH3, and GNP-PEG-biotin) distinguished the essential differences in drug binding efficiency and nanoconjugate stability.

The present study expanded the magnetic pH‐dependent, and nucleolin receptor‐targeted nano‐drug delivery system consists of synthesized superparamagnetic iron oxide nanoparticles (SPIONs) and poly N‐isopropyl acrylamide (PNIPAM) with aptamer AS1411 to deliver the erlotinib (ERL) to prostate cancer (PC) cells. Their properties were characterized using different techniques. The encapsulation efficiency (EE) and drug loading (DL) were found to be about 34% and 85%, respectively. The characterization studies disclosed that formulated SPION‐PNIPAM/ERL@AS1411 nanoparticles (NPs) were spherical with an average particle size of 97.8 nm, polydispersity index (PDI) of 0.03, and the zeta potential of −2 mV. The flow cytometry and cell viability assay results disclosed that SPION‐PNIPAM/ERL@AS1411 NPs operated cancer cell death via the pH‐sensitive release of ERL. The conjugation of the AS1411 modified the cellular uptake of ERL as proved by cellular sensing fluorescence. Therefore, the present study revealed that the SPION‐PNIPAM/ERL@AS1411 NPs improved the treatment effect of ERL in PC via the nucleolin‐targeted drug release.
