Recapitulating Trafficking of Nucleosides Into the Active Site of Polymerases of RNA Viruses: The Challenge and the Prize

Abstract: Nucleoside analogs are very effective antiviral agents with currently over 25 compounds approved for the therapy of viral infections. Still, their successful use against RNA viruses is very recent, despite RNA viruses comprising some of the most damaging human pathogens (e.g., Coronaviruses, Influenza viruses, or Flaviviridae such as dengue, Zika and hepatitis C viruses). The breakthrough came in 2013–2014, when the nucleoside analog Sofosbuvir became one of the cornerstones of current curative treatments for … Show more

“…In fact, computer simulations have been playing a key role in many biological systems including pharma and viruses (Barroso da Germain et al, 2011;Schlick and Portillo-Ledesma, 2021). Examples are found in several study cases from the understanding of capsid formation to the design of antiviral molecules (Boulard and Bressanelli, 2021;Francés-Monerris et al, 2020;Keretsu et al, 2020;Mondal and Warshel, 2020;Poveda-Cuevas et al, 2021Rapaport, 2018). Canonical simulation methods well established in more fundamental scientific areas and material sciences are now routinely used to enhance the understanding of the biomolecular interactions involved in infectious diseases.…”
