Recapitulating T cell infiltration in 3D psoriatic skin models for patient-specific drug testing

Abstract: Drug screening studies for inflammatory skin diseases are currently performed using model systems that only partially recapitulate human diseased skin. Here, we developed a new strategy to incorporate T cells into human 3D skin constructs (HSCs), which enabled us to closely monitor and quantitate T cell responses. We found that the epidermis promotes the activation and infiltration of T cells into the skin, and provides a directional cue for their selective migration towards the epidermis. We established a pso… Show more

“…As an example, we have reported the use of bioprinting technique to create a vascularized skin tissue [86] and the same approach can be applied to recreate a vascularized lung ALI tissue model. Addition of non-lymphocyte immune cells has also been explored using transwell plates with biofabricated tissue equivalents [87] which can be applied to the lung ALI assay systems to assess the participation of innate immune cells in infectivity and COVID-19 relevant immune responses. Transwell multi-well plates are compatible for laboratory automation equipment for the implementation of drugs screens.…”

“…The pathophysiology of psoriasis involves a complex interaction between keratinocytes and immune cells. Therefore, to accurately represent the disease condition, T cells were incorporated into 3D skin models [41,42]. These models were able to mimic the psoriatic epidermal phenotype as well as cytokine and transcription factor profiles.…”

“…Still, there is a need for psoriatic models, closely representing human disease, to be used as drug screening platforms. Recently, Shin et al (21) developed psoriatic human 3D skin constructs (pHSCs) by incorporating T cells, over the classical approach based on the use of patientderived keratinocytes or fibroblasts treated with PSO-related cytokines. As lymphocyte source, they tested in vitro polarized Th1/Th17 cells and CCR6 + CLA + T cells from the patients with PSO, both of which showed psoriatic phenotype on epidermal cells along with disease-associated cytokine profile.…”
