Realizing the Promise of Reverse Phase Protein Arrays for Clinical, Translational, and Basic Research: A Workshop Report

Abstract: Reverse phase protein array (RPPA) technology introduced a miniaturized "antigen-down" or "dot-blot" immunoassay suitable for quantifying the relative, semi-quantitative or quantitative (if a well-accepted reference standard exists) abundance of total protein levels and post-translational modifications across a variety of biological samples including cultured cells, tissues, and body fluids. The recent evolution of RPPA combined with more sophisticated sample handling, optical detection, quality control, and b… Show more

“…To gain insights into the molecular mechanisms that mediate the functional effects of PLAC8 knockdown in the cancer cells, we performed proteomic analyses of PLAC8 knockdown and control cells using RPPA technology (32). Whole cell lysates were probed with a total of 207 well-characterized antibodies examining expression levels and/or phosphorylation status of central regulators of cell growth or constituents of major signaling pathways.…”

“…Depending on the slide and pin size used to spot the protein lysates, a few hundred up to several thousand spots can be placed on each slide [34]. Lysates are usually spotted in a twofold dilution curve in duplicate or triplicate to allow exact quantification of the proteins of interest [53,54]. Therefore, detection of many biomarkers per sample is possible, allowing a detailed protein profiling of tissue samples.…”

“…To resolve this problem, one may use signal amplification strategy to improve the sensitivity. There are a variety of choices for signal amplification, such as TSA (Tyramide sginal amplification), dye precipitation, chemiluminescence, near-20 / 33 infrared dyes, quantum dots and ultra-sensitive planar waveguide technologies which are used in RPPA array (60). Another option is to enrich the amount of target protein from samples.…”
