Real-time near-infrared fluorescence imaging using cRGD-ZW800-1 for intraoperative visualization of multiple cancer types

Abstract: Incomplete resections and damage to critical structures increase morbidity and mortality of patients with cancer. Targeted intraoperative fluorescence imaging aids surgeons by providing real-time visualization of tumors and vital structures. This study evaluated the tumor-targeted zwitterionic near-infrared fluorescent peptide cRGD-ZW800-1 as tracer for intraoperative imaging of multiple cancer types. cRGD-ZW800-1 was validated in vitro on glioblastoma (U-87 MG) and colorectal (HT-29) cell lines. Subsequently,… Show more

“…Low dosages (equivalent to a total human dose of 0.096–1.33 mg) of cRGD-ZW800-1 were injected IV in rats. The study visualized the ureters 10-min post-injection up to a total duration of around 8 h. Toxicity studies of cRGD-ZW800-1 have been performed and report no adverse effects up to doses of 5.0 mg/kg in rats [ 28 ]. High SBRs of the ureter were found, peaking around 75.0, which is mostly due to the near absence of background fluorescence.…”

“…Smaller targeting agents -incorporating peptides, protein scaffolds, or antibody-fragments -with improved pharmacokinetic properties and specificity, have also been developed and are undergoing evaluation [25][26][27][28][29]. For example, fluorescent peptides have been clinically translated that were shown to improve the detection of polyps and neoplastic lesions in humans during an endoscopic examination at early time points [30][31].…”

“…There are a wide variety of peptides being investigated in laboratory settings including Arg-Gly-Asp (RGD), Somatostatin (SST), Gastrin releasing peptide (GRP) and Neurotensin (NT) among many others. In a recent study, a cyclic RGD-ZW800-1 was investigated as a generic tracer for multiple cancer types, and proved to be efficacious for both tumor and ureter identification [118]. The use of a zwitterionic dye such as ZW800 can overcome problems with background noise in FIGS.…”
