Real-Time Analysis of Mucosal Flora in Patients with Inflammatory Bowel Disease in India

Abstract: Mucosa-associated bacterial flora from control individuals and inflammatory bowel disease (IBD) patients were evaluated by real-time analysis using 16S rRNA-based genus-specific primers. Our data show a clear delineation in concentration of bacteria between the predominating and subdominating genera under disease conditions, indicating that the subsets of bacteria participating in the pathogenesis of ulcerative colitis (UC) and Crohn's disease (CD) are likely to be different.

“…47 In UC, the proportion of reads from Bacteroidetes phylum was higher than in health, consistent with a previous report, 32 but in contrast to others. 46,48 Although the proportion of reads from the phylum Firmicutes was lower, consistent with previous studies. 32,52 The lower reads in UC from the genera Coprococcus and Dorea have not been previously reported.…”

“…The increase of Firmicutes in IBD in European samples was previously observed,17 32 while others failed to find this pattern or even found the opposite 28 33 34. The Indian samples, in particular, showed either no or only weak signs of differential phylum abundance and diversity among disease conditions, by contrast with former lower coverage experiments in this ethnic group 35. These conflicting results might originate from the different sampling areas (here exclusively sigmoid colon) and types of samples (biopsies vs stool) in the respective studies.…”

“…Several of these have been linked to intestinal inflammation. [21][22][23][24][25][26] Of these, Prevotella, Desulfovibrio and an unknown genus in the Helicobacteriaceae family were almost only found in Room 2. Prevotella has the ability to degrade the sulfated mucin glycans, a modification that is more abundant in distal colon, which could affect the mucus layer.…”
