Reactivity of Biologically Important NSAID Compounds with Superoxide (O2.−), nitric oxide (.NO) and Cyclooxygenase Inhibition

Abstract: In the present study, we investigated the free radical-scavenging effects of eight Nonsteroidal Anti-Inflammatory Drugs (NSAID) using a Superoxide (O 2 .À ) and Nitric Oxide ( . NO) generating system in vitro. Indomethacine showed higher scavenging activity against O 2 .À , it being more potent than ibuprofen and nimesulide at the same concentration. Diclofenac showed a more relevant effect on the .NO scavenging activity than against O 2 .À . Comparison of the activities of eight compounds showed that the ar… Show more

“…It is important to emphasize that nitrate may also be formed, thus it should be reduced to nitrite prior to determination. For this purpose, NADH-dependent nitrate reductase was used for elimination of interference of NADH by addition of lactate dehydrogenase and pyruvate (Perez et al 2007). The chromophoric azo derivative formed from nitrite after Griess reaction is then measured spectrophotometrically at 548 nm.…”

“…For this situation, the nitric oxide staying after interaction with the sample is measured as nitrite. It is significant to confirm that nitrate can be formed, hence it ought to be diminished to nitrite before determination [ 35 ]. The derivative of azo chromophor was formed from nitrite after Griess reaction determined spectrophoto-metrically at 540 nm.…”
