Reaction of Porous Silicon with Both End-Functionalized Organic Compounds Bearing α-Bromo and ω-Carboxy Groups for Immobilization of Biomolecules

Abstract: Both end-functionalized (alpha-bromo and omega-carboxy) compounds were first tested for the radical reaction on the silicon-hydride (Si-H) terminated porous silicon (PSi) with/without the presence of diacyl peroxide initiator under microwave irradiation. Then the carboxylic acid monolayers (CAMs) assembled on PSi through the robust Si-C bonds were converted to amino-reactive linker, N-hydroxysuccinimide (NHS)-ester, terminated monolayers. And finally two proteins of bovine serum albumin (BSA) and lysozyme (Lys… Show more

“…The electrochemical etching was carried out in a 1:3 (V/V) pure ethanol and 40% aqueous HF for 12 min at a current density of 60 mA/cm 2 , similar to our previous report with minor improvement [14]. The freshly prepared PSi bears 5-10 nm nanometer pores, a porous layer thickness of ~1.5 m, and silicon hydride termini on the topmost surface.…”

“…Immobilization of biomolecules on the PSi surface has been reported by us and other groups recently [12][13][14][15]. Most widely-used immobilization strategies including physical adsorption, electrostatic and hydrophobic interactions, and chemical binding have been implanted onto the PSi surface [12][13][14][15][16][17][18].…”

“…Compared to that from curve a, the vibration frequency shifts due to the deeply associated OH. According to Beer's law, the amount of Si-H is proportional to its specific peak area in the absorbance mode of FTIR spectrum [28] . Since the water content is an important element for PFSA-based IPMC actuator, the complex absorbance area of OH···O is integrated to experientially evaluate the water content.…”

“…138 The latter methods have led to the immobilization of DNA, [138][139][140][141][142] peptides, 143 biotin-avidin sequences, [144][145][146] antibodies, 147 or other proteins. 129,148,149 The desired application then determines the functionality to be used. For example, Qin et al created an alkyne-functionalized monolayer, protected with a trimethylgermanyl group, which could be reacted subsequently with azide-containing mannose molecules in order to capture E. coli bacteria.…”
