RDM1 promotes critical processes in breast cancer tumorigenesis

Abstract: Breast cancer is currently among the most common cancers in women, with almost 200,000 new cases diagnosed annually. Dysregulation of DNA repair pathways allows cells to accumulate damage and eventually mutations, with a subsequent reduction in DNA repair capacity in breast tissue, leading to tumorigenesis. One component of the DNA damage repair pathway is RAD52 motif‐containing 1 (RDM1), but the specific role of RDM1 in breast cancer and the underlying mechanism remain unclear. Here, we examined the role play… Show more

“…As cancer progresses, more mutations accumulate in the DNA repair proteins of the tumor (41). Previous studies have shown that RDM1 potentiates RAD52-RAD51 signaling via p53-mediated transcriptional suppression (20,21). P53 plays an important role in regulating cell growth and apoptosis by triggering cell growth arrest or apoptosis through the regulation of various downstream targets (42).…”

“…Although there are few reports on the developmental role of RDM1 in tumors, studies have highlighted that RDM1 is upregulated in papillary thyroid cancer, non-small cell lung cancer, ovarian cancer and breast cancer. Knockdown of RDM1 can reduce the proliferation of tumor cells, increase cell apoptosis, and induce cell cycle arrest (18)(19)(20)(21). However, the role of RDM1 in LIHC remains largely unexplored.…”

“…In those cancers, RDM1 was regarded as an oncogene that could transcriptionally attenuate p53 expression and increase RAD51 and RAD52 level. This potential regulation of p53/RAD52/RAD51 signaling by increased RDM1 may lead to dysfunctional DNA repair pathways and the suppression of cell cycle arrest and apoptosis, thus promoting the tumor growth ( Hermeking and Eick, 1994 ; Tong et al, 2018 ; Chen et al, 2019 ).…”

“…Recently, some new works have revealed that RDM1 has high expression level in papillary thyroid carcinoma and lung adenocarcinoma [11][12][13]. However, the expression of RDM1 in ovarian cancer remains to be explored.…”
