RcsB Is Required for Inducible Acid Resistance in Escherichia coli and Acts at gadE-Dependent and -Independent Promoters

Abstract: RcsB interacts with GadE to mediate acid resistance in stationary-phase Escherichia coli K-12. We show here that RcsB is also required for inducible acid resistance in exponential phase and that it acts on promoters that are not GadE regulated. It is also required for acid resistance in E. coli O157:H7.The glutamate-dependent acid resistance system AR2 of Escherichia coli is regulated by a regulatory network that responds to general stress, via the alternative sigma factor RpoS, and to low pH (8,13,17,18,20,22… Show more

“…This set of differentially expressed genes is due most likely to the inactivation of the RcsB regulator in C ϩ variants of both RM6607 and RM6608. The roles of RcsB in curli production, acid resistance, cell motility, and biosynthesis of colanic acids and type I pili, etc., have been reported previously (20,30,35,56,67). The results of our study imply that RcsB may regulate heat shock resistance and cell catabolic activity, either directly or indirectly.…”

“…Elimination of Rfe, catalyzing a step in lipopolysaccharide (enterobacterial common antigen and O antigen) biosynthesis, and RcsB, which is the response regulator of a two-component system involved in activation of capsular colanic acid biosynthesis, likely results in an alteration of cell surface properties. RcsB is also required for EvgS/EvgA activation (71). Other gene products shown in Fig.…”

“…It has also been reported that RcsB, a response regulator of the RcsC/RcsD/RcsB TCS (in this case, a three-component system), is necessary for expression of GadE-dependent promoters of the AR genes (15). Furthermore, since EvgS/EvgA cannot be activated in a ⌬rcsB background, RcsB is essential for EvgS/EvgA activation (16). The RcsC/RcsD/RcsB system is known to be activated by various external stimulations, including low pH (17 EvgS is a hybrid histidine kinase composed of a histidine kinase domain, receiver domain, and Hpt transmitter domain.…”
