Rational design of a fluorescent receptor for the recognition of anthrax biomarker dipicolinate

Abstract: A new carbazole-2,7-dicarboxamide derivative has been synthesised and has been proved to effectively bind the dipicolinate anion, which is commonly used as an anthrax biomarker. The fluorescent response from this synthetic receptor offers a selective colour change in an organic-aqueous environment that is of valuable analytical use.

“…A flame-dried vial was charged with [4-(ethoxycarbonyl)phenyl]boronic acid 22 (123 mg, 0.634 mmol), (o-Tol) 3 P (11.6 mg, 0.038 mmol), and KF (73.6 mg, 1.27 mmol), then moved to a glovebox under a N 2 atmosphere. Pd 2 (dba) 3 •CHCl 3 (6.6 mg, 0.0064 mmol) was added to the vial, which was then capped and removed from the glovebox.…”

“…Its structural isomer isophthalate ( 12 ) was also selected; the meta ‐substitution provides a guest with a non‐linear arrangement of the two carboxylate groups. The dipicolinate guest ( 13 ) was selected for two reasons: (i) the structural similarity to the non‐linear isophthalate and (ii) the prevalence of this dicarboxylate in many bacterial endospores (a robust dormant form of bacteria) including anthrax , . As such the binding of this guest may be of biological significance.…”

“…For dipicolinate, while the modelled H:G arrangement was slightly “twisted” (Figure , bottom) the imide N–H was involved with an additional interaction to the pyridine N of the dipicolinate. Use of a central H‐bond donor in this manner has been successfully employed in the binding of dipicolinate by Curiel using a bispyrrole carbazole receptor …”
