Rat small intestine expresses the reelin–Disabled‐1 signalling pathway

Abstract: Expression of reelin, reelin receptors [apolipoprotein E receptor 2 (ApoER2) and very low density lipoprotein receptor (VldlR)] and the Disabled-1 (Dab1) protein was investigated in rat intestinal mucosa. Intestinal reelin and Dab1 mRNA levels were maximal in the early stages of life, reaching adult levels in 1-month-old rats. Expression of reelin mRNA was restricted to fibroblasts, whereas mRNAs of Dab1, ApoER2, VldlR and integrins α3 and β1 were observed in enterocytes, crypts and fibroblasts. Reelin protein… Show more

“…6 We previously proposed that in the rat small intestine the reelin released by the myofibroblasts might regulate the crypt-villus unit homeostasis 9 and the current data support this point of view. The results also show that the epithelial cell renewal processes vary with age, as previously reported for control mice, 15 and that reelin does not modify the pattern of this age-related changes.…”

“…9 Briefly, the small intestine was rapidly removed and washed with an ice-cold saline solution. One centimeter intestinal segments were incubated at room temperature in the phosphate-buffered saline (PBS) buffer containing 1 mM dithiothreitol for 15 min, followed by a 30-min incubation period at 37°C in a calciumand magnesium-free PBS buffer containing 1 mM EDTA and 2 mM glucose.…”
