Rat Corticotropin-Releasing Hormone Gene: Sequence and Tissue-Specific Expression*

Abstract: The rat corticotropin releasing hormone (CRH) gene has been isolated and characterized by DNA sequence analysis. The gene exhibits a structural organization similar to that of the human CRH gene. The nucleotide sequence encoding the entire rat CRH precursor is located on the second exon, while exon I encodes the 5'-untranslated region of the mRNA. Analysis of the nucleotide sequence homology between the human and rat CRH genes reveals several highly conserved regions including the CRH peptide-encoding sequence… Show more

“…TRH mRNA was localized to rat pituitary somatotropes (Bruhn 1994a,b,c) and TRH of pituitary origin can modulate both the secretion of TSH and the biosynthesis of pituitary TRH respectively (Bruhn et al 1998). Similarly, CRF mRNA was localized to the corticotropes in the rat pituitary (Thompson 1987, Giraldi & Cavagnini 1998 and there is evidence for the regulation of ACTH secretion by CRF of pituitary origin (Giraldi & Cavagnini 1998).…”

“…In situ hybridization histochemistry for CRF was performed on sections from both experiments 1 and 2 using a 48-mer cDNA oligoprobe corresponding to amino acids 22-37 of preprocorticotropin-releasing hormone (Thompson et al, 1987). In situ hybridization for AVP was performed on sections from experiment 2 using a 27-mer (AVP) directed against amino acids 110 -118 of rat prepropressophysin (Schmale et al, 1983).…”

“…Briefly, the CRF transgene was composed of the complete coding sequence of rat CRF cDNA (0.6 kb fragment; Thompson et al, 1987), which was inserted into a 8.2 kb genomic DNA fragment encompassing the murine Thy-1.2 gene, including regulatory regions and polyadenylation signal sequence (Aigner et al, 1995). The Thy-1 regulatory sequences drive constitutive transgene expression in postnatal and adult neurons (Morris and Grosveld, 1989;Vidal et al, 1990;Moechars et al, 1996;LĂŒthi et al, 1997;Wiessner et al, 1999).…”
