Rat bone marrow stem cells isolation and culture as a bone formative experimental system

Abstract: Bone marrow mesenchymal cells have been identified as a source of pluripotent stem cells with multipotential potential and differentiation in to the different cells types such as are osteoblast, chondroblast, adipoblast. In this research we describe pioneering experiment of tissue engineering in Bosnia and Herzegovina, of the isolation and differentiation rat bone marrow stromal cells in to the osteoblast cells lineages. Rat bone marrow stromal cells were isolated by method described by Maniatopulos using thei… Show more

“…On the third day, half the volume of the culture medium was changed and fresh medium were replaced every 3 days, nonadherent cells (haematopoietic stem cells and red blood cells, etc.) were eliminated through medium changes, 8,10 while the BMSCs can adhere to plastic surface and easily expand during in vitro culture. 9 The BMSCs which were grown to 80% confluence were defined as passage zero (P0) cells.…”

“…8 Bone marrow mesenchymal stem cells can adhere to a surface of a plastic flask, 9 and haematopoietic cells remained suspended in the medium, and they were mostly eliminated through subsequent medium changes. 10 Therefore, we used direct adherence to isolate adherently growing BMSCs from whole marrow cells. Bone marrow mesenchymal stem cell purity increases with medium changes; passage 8 (P8) BMSCs display an obvious aging trend, and P3-5 cells have high purity.…”

“…Bone marrow derived MSCs were obtained on the basis of their ability to adhere to the culture plates as described elsewhere [23]. Briefly, bone marrow cells from deep anesthetized Wistar male rats (8 -10 weeks old) were collected by flushing the femurs and tibias and were rinsed twice by centrifugation at 1,200 rpm for 5 minutes in Phosphatebuffered saline (PBS).…”
