Rapidly Increasing Serum 25(OH)D Boosts the Immune System, against Infections—Sepsis and COVID-19

Abstract: Vitamin D deficiency is a global public health problem, a pandemic that commonly affects the elderly and those with comorbidities such as obesity, diabetes, hypertension, respiratory disorders, recurrent infections, immune deficiency, and malignancies, as well as ethnic minorities living in temperate countries. The same groups were worst affected by COVID-19. Since vitamin D deficiency weakens the immune system, it increases the risk of infections, complications, and deaths, such as from sepsis and COVID-19. D… Show more

“…However, this picomolar calcitriol concentration in the circulation is insufficient to enter extra-renal target cells. Therefore, most non-musculoskeletal functions, as with autocrine and paracrine signaling of immune cells, are driven by a three-orders of the magnitude of calcitriol concentration generated within these cells [30,31].…”

“…Therefore, regardless of the size or expenses of the RCT, the reported failures of vitamin D trials [66] were not due to a lack of effect but due to flawed study designs [31], in conjunction with failure to adhere to Heaney's criteria for causality in a biological system [67]. Consequently, those studies report negative results and misleading conclusions because of researchers' failure to understand vitamin D's biology and physiology.…”

“…Importantly, not only does the treatment of cell culture models with 1,25(OH) 2 D 3 promote changes in gene expression, but also the supplementation of individuals with vitamin D 3 leads to the same results. Most of these studies were conducted on patients with diverse inflammatory diseases, such as COVID-19, colorectal cancer, irritable bowel syndrome, obesity and diabetes [ 28 , 29 , 30 , 31 , 32 , 33 , 34 ], and only a few studies were performed with healthy individuals [ 35 , 36 , 37 ].…”
