“…These are, however, attempts which accelerate individual steps of the DNA analysis process. The RapidHIT TM 200 from IntegenX Inc., in contrast, offers an integrated single-instrument platform that combines sample DNA extraction, STR-PCR amplification, amplicon separation and detection, and DNA profile analysis, all within a relatively short duration of about 2 h [16][17][18][19][20][21][22].…”

“…The instrument has been manufacturervalidated [16][17][18] and tested by various groups [19][20][21][22] for the processing of reference buccal samples. In the present study, we seek to evaluate the performance of the RapidHIT TM 200 to generate DNA profiles from samples routinely encountered in crime casework.…”

“…In the United States, police agencies in Arizona, Florida, and South Carolina, to name a few, have also begun using this platform for the generation of investigative leads for casework samples. The rate of DNA profiling success for casework samples appears to be sample-dependent, but full profiles have been generated from cigarette butts (range of success 0e100%, n ¼ 29 samples from 6 donors); drinking items (range of success 6e100%, n ¼ 13 samples from 10 donors); and chewing gum (range of success 0e100%, n ¼ 23 samples from 16 donors) (Verheij et al, 2013). Given this high variability in success rates for profile detection with forensic type samples, additional improvements in the sensitivity of these systems will be needed to allow for their application to the analysis of more challenging sample types, such as touch or contact DNA samples, which typically have only trace quantities of DNA.…”

“…When DNA quantities are not as limiting as with touch type samples, it has been demonstrated that it is possible to produce full DNA profiles using traditional laboratory methodologies following a reextraction of samples previously analyzed by the RapidHIT Ô system. This indicated that the RapidHIT Ô system does not consume all available biological material (Thong et al, 2015;Verheij et al, 2013). This is important considering the limited nature of many forensic type samples, since it demonstrates that if further efficiencies in DNA extraction can be achieved, the success rate with casework samples might be improved.…”

“…The RapidHIT system is capable of producing STR profiles in less than 90 min with several advantages of full integration, automation, simplification, and no human intervention. The RapidHIT system contains two cartridges with eight slots (for five samples, one positive control, one negative control, and one ladder), a sample pretreatment system for bead‐based DNA isolation, a PCR amplification system with use of GlobalFiler Express Kit or PowerPlex 16 HS Kit, and a CE system using a fused‐silica capillary array .…”
